grjj:张翱,博士、中国科学院上海药物所研究员、博士生导师、国家****基金获得者和上海“浦江人才计划”优秀奖获得者;曾先后在Georgetown University Medical Center和Harvard Medical School- McLean Hospital进行博士后研究,并于2004年正式成为Harvard Medical School的Professional Investigator, 2004年开始担任Harvard Medical School的Instructor及药物化学研究室助理主任(Assistant Director)。主要从事化学生物学及药物化学研究,尤其是针对神经精神性与肿瘤两大疑难疾病,瞄准在药物研究中具有重要意义和挑战性的G-蛋白偶联受体(GPCR)和受体酪氨酸激酶(RTK)为靶点,系统地开展了基于类药性优势骨架的高效合成、结构优化和聚焦化合物库的构建研究,获得了百余个活性化合物,其中:针对现有中枢镇痛药物的高成瘾性缺点设计的κ-受体激动剂ATPM-ET,已作为新型低成瘾性镇痛药物进入系统临床前研究;针对选择性多巴胺受体激动剂抗帕金森药物易产生异动症的缺点,率先在国际上开展了同时具有D2和5-HT1A受体双重作用机制药物研究,获得的双重全部激动剂化合物MCL-135,作为低毒有效的新型抗帕金森候选药物,正在进行早期临床前研究;在国际上首次完成天然产物Marmycin A的基本骨架的合成,并通过嵌入激酶抑制剂优势药效团构建了咔唑类优势骨架化合物库,获得了一系列靶向酪氨酸激酶c-Met 特异性抑制剂,同时还获得PARP1高活性抑制剂,体内体外均具有较强的抗肿瘤活性,目前已进入临床前系统性药效和安全性评价,极具开发前景。已发表SCI论文90余篇,其中在药物化学国际一级期刊Journal of Medicinal Chemistry 发表论文12篇,以第一申请人申请国内国际专利35项。先后获邀为Chem Rev、Med Res Rev、J Med Chem等就药物发现新策略新理念和相关药物研发进展撰写评述和展望,并担任许多国际期刊的审稿人。曾获2004年Harvard Medical School年度最高学术奖Alfred Pope Award及Adam Cornneel青年奖并获得该基金资助。主持的中科院和上海市“浦江人才计划”结题均获优秀, 2011年获国家****基金。主持完成“十一五”国家重大新药创制重大专项课题、国家自然基金等课题;正在主持 “十二五”国家重大新药创制重大专项课题、国家“****”课题等重要项目。
