grjj:戴辉雄博士现任中国科学院上海药物研究所研究员、课题组长。2007年在中科院上海有机化学研究所获得博士学位,导师:陆熙炎院士。2007-2009年,美国德州大学从事博士后研究,导师;H. V. Rasika Dias教授;2009-2012年,美国Scripps研究所担任研究助理,导师:余金权教授;2012.7-2016.2中国科学院上海有机化学研究所担任副研究员; 2016.2-至今,中科院上海药物所担任研究员。目前主要致力于开发新型的官能团转化方法,用于天然产物和药物分子的结构修饰,从而发展出高效的先导化合物的优化策略,更好地服务于新药研发。具体研究方向为C-H键和C-C键活化官能团化。在国际学术期刊Nature,J. Am. Chem. Soc, Angew. Chem. Int. Ed.等上已经发表论文40余篇,他引达到了2600余次;申请专利2项。

kycg: 1.围绕C-H键活化,以金属钯、钌以及铜作为催化剂,发展了一系列官能团转化新方法,构建新的C-C, C-CF3, C-N, C-O, C-B键;发展了两类全新的导向基团, 一类为药物中广泛存在的磺酰胺, 另一类为酰胺-噁唑啉(amide-oxazoline);成功实现了一类由低价金属出发的活化体系,通过对芳基和杂芳基C-H键活化构建亚氨基异吲哚啉酮类化合物以及对烯基的α和β-C-H键选择性活化合成β和?-亚氨基内酰胺类化合物,同时也很好地克服了杂环在这类碳氢键活化中的不利影响。

2.发展了一类配体促进的C-C键活化新策略,实现了芳基酮类化合物的多样性转化。

3.药物化学中的应用:1)将“酰胺-噁唑啉”导向基团策略运用到含杂环的复杂药物分子修饰中,实现了对抗高血压药Telmisartan的选择性官能团化:高选择性地实现了在含多个杂环的复杂分子体系中的羧酸邻位胺化、羟基化、硫醚化、芳基化以及三氟甲基化等六个转化,快速在药物分子Telmisartan上引入了各种官能团;2)通过一类新型C-H键活化/卡宾插入/Lossen重排串联反应,高效构建了一类含吲哚酮的螺环骨架。
