本实验室利用分子生物学手段研究与植物叶绿体发育相关的代谢过程及其调控机制。
牻牛儿基牻牛儿基二磷酸(GGPP)是植物萜类代谢途径中的一个关键分支点。植物以GGPP为底物,合成大约2万种天然产物,其中包括激素(赤霉素、脱落酸、独脚金内酯)、色素(类胡萝卜素和叶绿素的植基侧链)、维生素(来自胡萝卜素的维生素A和维生素E的植基侧链),以及参与植物对病原微生物防御机制的二萜植保素。这些代谢分支共同竞争GGPP,构成了牵一发而动全身的复杂网络。GGPP的合成及其在下游分支间的分配机制影响了植物的生长发育和逆境适应能力,也影响着农产品的营养品质和风味特色,以及园艺植物的观赏价值。
本实验室近年来围绕这一核心问题开展了一系列研究工作。主要进展包括:
1. 解析了水稻中GGPP保障叶绿素代谢的调控机制(Zhou et al., 2017, PNAS);
[类囊体膜上LIL3作为锚蛋白结合GGPPS及其招募蛋白GRP,将代谢流引入叶绿素合成途径(经叶绿素合酶CHLG催化)。我们发现并解析了GGPPS/GRP异源二聚体和GGPPS同源二聚体,并鉴定了其酶促动力学特性。]
2. 解析了锌指蛋白ORANGE在子叶中通过转录因子TCP14对叶绿素合成和叶绿体发育的抑制作用(Sun et al., 2019, Plant Cell);
[TCP14在细胞核内诱导ELIPs基因从而促进叶绿素合成与叶绿体发育。在黑暗中OR蛋白位于核里并抑制TCP14。种子破土而出后,核里的OR蛋白逐渐减少,从而推动叶绿素合成与叶绿体发育。而转入叶绿体的OR则促进类胡萝卜素合成来形成对叶绿体新的保护]
3. 解析了辣椒果实中类胡萝卜素合成的部分代谢流调控机制(Wang et al., 2018, 219, J Agric Food Chem);
4. 以紫菜为主要材料,系统研究了红藻的类胡萝卜素代谢途径,初步揭示了这一重要代谢途径在植物中的早期演化(Yang et al., 2014, J Intergr Plant Biol; 2015, J Appl Phycol; Cao et al., 2017, Mar Drugs; Deng et al., 2020, J Agric Food Chem);并建立了基于蓝细菌的酶活鉴定系统和合成生物学光驱动底盘(Cao et al., 2020, Plant Biotechnol J)。
