My main research focus is on diabetes and obesity in adults and the youth populations. Key aspects include epidemiology and pathogenetic mechanisms, in particular, sleep as a novel lifestyle risk factor and its interaction with diet, in mediating the development of obesity and obesity associated cardiovascular risk factors, as well as influencing glycemic control in patients with type 2 diabetes.
Nearly two decades ago, I set out to conduct clinical trials involving Troglitazone, the first thiazolidinediones (TZD) as insulin sensitizer for the treatment of type 2 diabetes (Diabetes 2002). TZD is a pharmacological peroxisome proliferator-activated receptor (PPAR) gamma agonists which cause fat redistribution with reduced free fatty acids production, improved insulin resistance and reduced inflammatory responses. I have been running randomized control trials including new pharmacological agents such as new PPAR gamma agonist, Rivoglitazone (Diabetes Obesity Metabolism 2011). In addition, I conduct clinical trials exploring the role of various lifestyle factors including low glycemic index diet (Obesity Review 2011) and sleep (Sleep Medicine 2011) in modulating the development and disease course of patients with obesity and diabetes.
As the Steering Committee Member of Joint Asia Diabetes Evaluation (JADE) programme, an internet-based registry for patients with type 2 diabetes in Asia countries and cities, my current programme is also focused on the improvement of disease management of patients with obesity and type 2 diabetes with an emphasis on protocol-driven model (Diabetes Care 2007) and personalized care targeting on the unmet needs of patients with type 2 diabetes in the East (Nature Review Endocrinology 2013).
