摘要BRE (Brain and Reproductive Expressed) is a highly conserved, putative stress-modulating gene. It has been shown able to bind the cytoplasmic region of p55 tumor necrosis factor receptor 1 (TNFR-1), whose activation through binding with its ligand, TNF-alpha, triggers both the apoptosis and pro-survival pathways, the latter of which is mediated by activation of NF-kappaB. Overexpression of BRE has also been shown inhibitory to the activation of NF-kappaB by TNF-alpha. This, we surmised, would skew the effect of TNF-alpha towards apoptosis. In this study, we investigated the functional consequence of BRE overexpression on TNF-induced apoptosis, and sought to confirm the above prediction. Our data, however, demonstrated the opposite result. Using both the human and mouse cell lines, HeLa and D122, respectively, we found that overexpression of BRE in these cells through stable transfection with the BRE expression construct, pcDNA3-BRE, resulted in resistance to TNF-alpha-induced apoptosis, in the presence of cycloheximide. Thus, we have shown that despite the reported ability of BRE in down-regulating the survival-promoting NF-kappaB activity induced by TNF-alpha, BRE can inhibit the apoptosis induced by the same cytokine.
