DNA甲基化和组蛋白修饰引起的染色质变化对于基因组稳定性、基因表达调控、基因组印记和转基因沉默具有重要作用。DNA甲基化是指在DNA的胞嘧啶碱基的5号位加上一个甲基,它广泛存在于原核生物和真核生物中。在植物基因组中,甲基化的DNA位点往往伴随产生小分子的siRNA和非编码的长链RNA,这些RNA可以通过RNA指导的DNA甲基化途径引导基因组DNA的甲基化。组蛋白修饰包括甲基化、乙酰化、磷酸化和泛素化等,这些修饰和DNA甲基化协同作用,共同调控染色质的状态。我们将利用模式植物拟南芥为研究材料,通过利用遗传、分子和生化方法重点研究RNA指导的DNA甲基化和组蛋白修饰途径。我们将鉴定这个途径中的新组分,并研究它们在维持基因组稳定性,调控发育和转基因沉默机制中的重要功能。这将有助于我们深入理解植物体在发育和环境适应过程中表观遗传的动态调控机制。ResearchDescriptionChromatinchangescausedbyDNAmethylationandhistonemodificationsplaycrucialrolesingenomestability,generegulation,genomicimprintingandtransgenesilencing.DNAmethylationreferstoadditionofamethylgroupto5’-cytosineofDNA,whichextensivelyexistsfromprokaryotestoeukaryotes.Inplants,themethylatedgenomicDNAlociarefrequentlyaccompaniedbytheoccurrenceofsiRNAsandnoncodingRNAs,whichcausedenovoDNAmethylationthroughRNA-directedDNAmethylation.Thehistonemodificationsincludemethylation,acetylation,phosphorylationandubiquitination,whichcoordinatewithDNAmethylationandregulatechromatinstatus.WewillstudytheRNA-directedDNAmethylationandhistonemodificationmechanismsbyapplyinggenetic,molecularbiologicalandbiochemicalapproaches.Weattempttoidentifynewcomponentsinthepathwayandstudytheirfunctionsingenomestability,developmentandtranscriptionalgenesilencing.Thesestudieswillhelptounderstandthedynamicregulationofepigenomesduringplantdevelopmentandadaptationtoenvironments.发表文章Publications1.HeXJ,HsuYF,ZhuS,LiuHL,PontesO,ZhuJ,CuiX,WangCS,ZhuJK.(2009)AconservedtranscriptionalregulatorisrequiredforRNA-directedDNAmethylationandplantdevelopment.GenesDev.23,2717-2722.
