研究概述本实验室致力于细胞电子断层成像技术的开拓及其在生物医学研究中的应用。细胞电子断层成像技术是一项新兴的生物电镜三维成像技术,它可以在纳米尺度解析瞬时冷冻固定的细胞中的精细结构、动态事件及分子定位,它有机地整合了生物细胞组织样品的超快速冷冻固定、冰冻切片、低温去水固定、基因技术、蛋白质及基因的标记识别、冰冻电镜成像和计算机图像处理等技术(见图1)。结合我们最新开发的蛋白质、基因标记的电镜识别定位技术,本实验室将开展一系列生物医学相关课题的细胞电子断层成像技术研究。目前本实验室的主要研究方向如下:1.生理状态下活体细胞中的蛋白质、基因的标记及相应的电镜检测定位技术
2.瞬时捕捉生物体中动态分子事件和纳米尺度解析细胞的精细结构及复合体的组装过程
3.解析疾病状态下(如病原体和宿主细胞的相互作用过程中的)动态分子事件及相应细胞精细结构的变化
4.纳米尺度精确定位跟踪某些关键分子在胚胎发育及干细胞分化过程中的时间空间分布
ResearchDescriptionOurmajorinterestisfocusedonthedevelopmentofcellularelectrontomographyplatformtechnologyanditsapplicationsinbiomedicalresearch.Cellularelectrontomographyisanemergingthree-dimensionalelectronmicroscopybio-imagingtechnology,whichcanbebroadlyusedtodissectthecellularultrastructures,dynamicevents,andpreciselylocatingindividualmoleculeintherapid-frozencellsatnanometerresolution.Cellularelectrontomographyplatformtechnologyinvolvesrapid-freezingtechnique,frozen-hydratedthinsectioningtechnique,freeze-substitutionfixation,genetechnology,proteinlabeling/genetictaggingandthecorrespondingdetectiontechniques,cryo-electronmicroscopyandcomputerimagingprocessingtechniques.Wewillinitiateaseriesofbiomedical-relatedcellularelectrontomographyprojectswithournewlydevelopedproteinlabeling/genetictaggingandthecorrespondingdetectiontechniques.Ourcurrentresearchwillfocusonthefollowingareas:1.Developingnovelinvivoproteinlabeling/genetictaggingandthecorrespondingelectronmicroscopydetectiontechnologies;
2.Dissectingthedynamicmolecularevents,cellularultrastructuresandtheassemblingprocessesoflargeproteincomplexesinrapid-immobilizedcells;
3.Untanglingthedynamicmoleculareventsandtheultrastructureschangesthatinvolveindiseases,suchashost-microbeinteractions;
4.Tracingtheprecisetemporal-spatialdistributionsofthekeyproteinsinvolvedinembryonicdevelopmentandstemcelldifferentiationprocesses.
