实验室概况 Our Lab
实验室团队现有教授1名(孙少琛),讲师1名(张羽),博士生4名,硕士研究生9名。实验室研究生有12人次获得校长奖学金、国家优秀研究生奖学金;1人次获得江苏省优秀博士学位论文;3人次获校优秀博/硕士学位论文;1人次获南农好榜样荣誉;3人次获国际会议优秀海报奖;研究生前往泰国、越南等地参加国际会议,并做会议报告3人次。实验室承担本科2年级国家级SRT项目、本科4年级毕业设计等多项,指导本科生获得校优秀毕业设计特等奖、一等奖各1项。
实验室在Development、Hum Reprod、Biol Reprod、Archi Toxicol 等学科权威期刊已发表SCI 论文100多篇,其中第一/通讯作者SCI 论文70多篇。研究成果在Cell Cycle、Mol Hum Reprod、Biol Cell等获得编辑和同行专家特邀评论、8篇研究论文当选为当期杂志封面文章,被多本英文著作如Oogenesis收录并介绍,并做WCRB,ARBS等多个国际大会特邀报告。
研究生招生就业情况:实验室学术型与专业型硕士均招收,方向偏重基础研究,毕业生倾向于医院和高校就业。毕业博士/出站博士后共7人(5人高校工作,2人事业单位工作);已毕业硕士13人(8人英国澳洲韩国等攻读博士学位,5人企事业单位)。
研究概况 Our Research
本课题组的研究重点在哺乳动物卵子质量的影响因素和分子调控机制。卵子质量的下降会引起受精或者胚胎发育失败,直接关系到动物生产的经济效益,也是导致人类生殖障碍的重要因素。影响卵子质量的因素主要分为内部和外部两个方面。内部因素主要是卵子成熟的自身分子调控,包括细胞周期、细胞骨架及线粒体功能等生物学时间;外部因素主要是环境因素影响,包括饲料毒性、温度、应激等。本实验室具体的研究方向为:
1)卵母细胞成熟过程中细胞骨架的分子调控
卵母细胞生发泡破裂后,染色质凝集成染色体,微管在附近组装形成纺锤体并牵引染色体在中央排列。微丝推动纺锤体/染色体迁移到卵母细胞的皮层下,染色体分离,通过不均等分裂形成一个大的卵子和一个小的极体。纺锤体组装与染色体分离保证了遗传物质的稳定和准确性,微丝介导的不均等分裂为受精和早期胚胎发育保留母系物质。因此,探清微丝对卵子成熟的分子调控机制对于卵子质量控制和预防生殖缺陷是非常重要的。
课题组的工作证明GTP酶家族RhoA主导的ROCK-LIMK-Cofilin、以及微丝聚合因子家族NPFs-Arp2/3信号通路调控微丝骨架动力学变化参与卵母细胞中微丝介导的纺锤体迁移和胞质分裂,Arp2/3通路已被收录入Oogenesis等英文著作中。我们的结果揭示了GTP酶家族、微丝聚合因子等两大类微丝骨架调控因子在卵子中的作用,提出了控制卵子不均等分裂的Arp2/3和RhoA主导的两条信号通路。
2)环境对卵子质量的危害及机制
近年来国内食品环境安全问题频发,女性生殖障碍相关问题日益增多;在家畜养殖中发霉饲料中霉菌毒素则造成家畜流产死胎等现象,极大影响了家畜生产的经济效益。对于人类及家畜出现的各种繁殖问题,卵子质量下降是重要原因之一。研究环境毒素对于卵子质量的影响,有利于为防治这一问题提供理论基础。
近期实验室采用体内/体外模型,报道了摄食含有霉菌毒素饲料的小鼠和猪的卵子质量下降,发育能力、亚细胞结构分布和定位均发生异常。我们证明霉菌毒素能够诱发卵子氧化应激,从而导致卵子发生细胞凋亡和自噬; 此外,霉菌毒素处理改变了卵子DNA和组蛋白甲基化的水平可能是霉菌毒素对卵子毒性的另外一个原因。此外,褪黑素的添加能够有效缓解氧化应激水平,挽救霉菌毒素造成的毒性损伤。实验室同时也研究三聚氰胺,丙烯酰胺,双酚A等环境污染物对卵子的毒性损伤。
