简介:

1964年4月出生,华南师范大学生物光子学研究院院长、学术委员会主任,激光生命科学教育部重点实验室教授,博士生导师,广东省“千百十人才工程”省级培养对象。主持国家863项目1项(230万)、国家重大科研仪器研制项目1项(510万)、国家自然科学基金面上项目5项,主持广东科技计划项目1项、重点项目1项以及其他省部级项目4项。研究领域主要涉及生物医学光子学、生物物理学、肿瘤细胞生物学、分子生物学。发表学术论文140余篇,获得授权国家发明专利5项。

(一)荧光共振能量转移(FRET)技术简介
FRET(Fluorescence/Ferster resonance energy transfer)是一个依赖于供体和受体分子间距离的光物理进程,处于激发态的荧光团通过偶极子间的相互作用将能量以非辐射的方式转移给邻近的受体分子,从而导致供体荧光淬灭和受体荧光发射的增加。供体和受体之间的能量转移效率 (E) 与分子间的空间距离 (r) 满足6次方的关系:

现代生物学的研究不仅需要了解参与生物过程的分子种类,更需要了解生物学过程的精确分子调控机制。尽管体外和体内研究已经揭示了大量特异的生物分子之间的相互作用,但大量发生于生物体内动态的或瞬时性的或者弱的分子间相互作用目前还难以准确鉴定和深入研究
荧光共振能量转移(FRET)显微成像技术是目前唯一(unique)可以在单个活细胞中直接实时高时空分辨检测分子间动态相互作用过程的成像技术。基于荧光蛋白(FPs)FPs的FRET显微成像技术(FPs-FRET)已经成为在单个活细胞中研究分子间动态相互作用的最主要技术手段。
(二)本课题组开展对外服务
本课题组发展了多种单个活细胞中FRET效率的定量检测技术,获得多项国家发明专利,目前可开展多种形式的对外合作服务。
1. 合作形式:
(1) 独立承担单个活细胞中FRET定量检测分析;
(2) 联合申报国家和省部级重大科研项目。
2. 研究问题类型:
(1) 在单个活细胞中研究蛋白质间相互作用;
(2) 在单个活细胞中研究蛋白质寡聚化;
(3) 在单个活细胞中检测信号转导分子间的动态调控过程;
(4) 在单个活细胞中研究蛋白激酶的活性;
(5) 在单个活细胞中研究蛋白质的磷酸化;
(6) 在单个活细胞中研究生物大分子的构象变化。
