研究概述成体干细胞或组织特异干细胞长期存在于多种组织和器官内,对维持机体的动态平衡及组织的再生或修复能力起决定作用。干细胞本身需要接受严格的调控,调控的紊乱可能导致癌症,衰老或退行性疾病的发生。我们实验室致力于研究组织和器官的成体干细胞在体内是如何长期维持的,它们处在一个什么样的微环境内,如何应对环境的改变,它们基因缺陷如何导致了组织和器官功能的改变和疾病的发生。在过去几年的研究中,我们在果蝇干细胞的微环境结构,维系干细胞的信号因子及内源性的表观遗传机制等方面有了开创性的发现。这些发现为进一步理解干细胞的干性在遗传和表观遗传方面的分子机制提供了基础。另外,我们正在建立和研究原位干细胞突变致瘤和组织再生的果蝇及小鼠模型。这些研究将对肿瘤等疾病的发生机制和防治、体外成体干细胞扩增和干细胞治疗提供理论依据和实验基础。ResearchDescriptionAdultstemcellsornamedtissue-specificstemcellsresideinmanytissueandorgansandhavecriticalrolesinmaintainingnormaltissuehomeostasisandregenerationthroughoutlifeofeachorganism.Theirdysfunctionisbelievedtobeintimatelyassociatedwithvarioustypesofdiseasessuchascancer,degenerativediseases,andageing,buttheunderlyingmechanismsarepoorlyunderstood.Mylaboratoryisinterestedinunderstandinghowstemcellsaremaintainedwithineachtissue,howdotheyrespondtoenvironment,andhowtheirdysfunctionleadstodiseases.Overthepastseveralyears,wehavemadeimportantprogressonunderstandingthenichestructure,maintenancesignals,andintrinsicself-renewalmechanismsforDrosophilastemcells.Thesestudiesprovidethefoundationforfurtherunderstandingtheregulatorymechanismsofstemcellself-renewalatbothgeneticandepigeneticlevels.Inthenearfuture,wewillcontinuetodissectthemolecularandgeneticprogramsunderlyingstemcellself-renewalinDrosophila,withanewemphasisontheestablishmentandgeneticdissectionofstemcell-originatedtumorigenesisaswellasdamage-inducedtissueregenerationmodelsinbothDrosophilaandmouse.Publications:1.SunP,QuanZ,Zhang,B,WuT,XiR(2010).TSC1/2tumoursuppressorcomplexmaintainsDrosophilagermlinestemcellsbypreventingdifferentiation.Development137:2461-2469.
