课题组简介
仿生体系的分子组装就是在分子水平或者说纳米尺度上,采用化学和物理的方法,将分子生物学与纳米工程相结合,模仿生物体特定的组成、结构、状态和功能,设计和构造新型的具有生物功能的体系、材料或器件。本课题组致力于分子组装和纳米结构体系设计的研究,尤其是基于生物分子的仿生体系组装与物理化学机理的研究。以模拟生物体结构和功能为基础,在分子水平上构建具有特殊功能的分子组装体,旋转马达蛋白与线性蛋白的重组,寡肽分子自组装及结构转变过程调控,以及发展模板组装技术构建系列功能化纳米结构体系并应用于外缘物质的胞内输送。

1、超高分辨荧光显微镜
超高分辨荧光显微镜是近年来兴起的新技术,它可以超越远场光学显微镜的分辨率极限,即阿贝极限(200 nm左右)的限制,直接检测到几十纳米的精细结构。与能达到相同或更高分辨率的X光显微镜,各类电子显微镜及原子力显微镜相比,超高分辨荧光成像的优势是在常温常压和基本不损伤生物样本活性的条件下,获得纳米尺度的图像信息。
我们课题组在超高分辨结构图像采集和数据分析方面发展了实时单分子定位的程序包AutoBayes,并将其应用于高背景的细胞成像数据分析(Sci. Rep., 2015, 5, 11073.),蛋白质在生物矿物中的分布(Angew. Chem. Int. Ed., 2016, 55, 908-911),外源脂质体或组装体在细胞骨架上的分布和运动轨迹的实时检测(Sci. Rep., 2015, 5, 16559)。

2.1 旋转马达(CFoF1-ATPase) ——仿生合成ATP
ATP 合酶是生物体内能量代谢的关键酶,也是自然界中最小的生物分子马达,具有从生物能量的产生到纳米器件的构筑等一系列潜在的应用价值,成为当前世界许多领域的研究热点。基于这一背景,我们致力于将ATP 合酶重构在具有特定尺寸、组成、性质以及功能的微胶囊/微球外层,研究ATP合酶的体外重组、功能化以及在仿生合成ATP中的应用前景。(Angew. Chem. Int. 近年来,肽分子的自组装由于具有分子设计灵活、组装结构多样以及功能可调等优势而成为分子仿生体系的研究热点,并在药物输送、基因转染、组织工程以及生物传感等诸多领域得到广泛应用。我们课题组以芳香类线性二肽-二苯丙氨酸及其衍生物作为构筑基元,在不同的实验条件下动态调控肽分子的自组装过程,获得了一系列具有一维、二维以及三维形貌的纳米组装结构。通过改变组装体系的浓度、变换溶剂以及引入外源小分子诱导等方式成功实现了二苯丙氨酸基肽分子多功能,多级结构的可控自组装。Adv. 在分子水平上组装多功能仿生微胶囊/微球引起了科学家们的极大兴趣。由于层层组装技术简单易行,组装材料来源丰富,组装形貌精确可控等优势,使得其成为制备仿生微纳米结构的最佳途径之一。我们课题组选用生物大分子(如多糖和蛋白质等)作为组装基元,利用层层组装技术与模板法相结合制备了一系列仿生微胶囊/微球/纳米管,并成功将这些仿生微纳米结构应用于癌症治疗、血液代用品等生物医学领域中。Chem. 1992年德国美因兹大学物理化学研究所联合培养博士生。
1992年12月于吉林大学化学系获博士学位。
1993年至1996年先后在希腊国家研究与工程基地、德国马普胶体与界面研究所从事博士后研究。
1996年中国科学院原感光化学研究所研究员、博士生导师。
1999年中科院化学所研究员、博士生导师。

Molecular assemblies of biomimetic systems: Membrane hydrolysis, complex assembly of lipids, proteins, enzymes and biomolecular motors: F0F1, Kinesin, Actin, biogenic microcapsules, self assembly and in vitro characterizations of biological molecules such as DNA, peptide and single cells as well as their mixtures with surfactants and polymers.

Biointerfaces: molecular patterns, surface modification, molecular recognition of enzymes, chemical recognition at cell surfaces, transport through cell membranes, design and synthesis of bioinspired molecules and materials for drug release and gene delivery.

Nanostructures: Design, synthesis, characterization and functionalization of nanoparticles, nanopatterns, nanotubes, nanocrystals.

Tunable Self-Assembly of Dipeptide : tunable self-assembly, tunable disassembly, structural transformation, design, synthesis, characterization and functionalization of dipeptide assembly structure.
