综合介绍 General Introduction
戴博博士是西北工业大学医学研究院的副教授。他早年追随动物胚胎工程专家张涌院士学习克隆羊技术,取得硕士学位。之后赴美国留学,进行人胚胎干细胞的研究,2009年毕业于康涅狄格大学,获得哲学博士学位。2010年进入斯坦福大学医学院,接受博士后培训,师从著名的血液学家Ravi Majeti教授及Irv Weissman院士,从事人类早期血液发生机理及白血病干细胞的研究。归国以后,领导了陕西省干细胞应用工程研究中心,开展干细胞相关的研究和转化工作,先后入选了“西安市引进海外高层次创新人才”和“ 陕西省高层次人才”。兼任西安医学院客座教授、国际干细胞学会会员、(美国国家心肺血研究所)祖细胞生物学联盟会员、美国《觉悟视频科学杂志》终身评委会委员,及多家国际学术期刊特约审稿人。
戴博博士有近二十年的干细胞研究经验。攻读博士期间,创新性地在国际上提出“表观蛋白质组”的概念,并被2012年诺贝尔生理学或医学奖得主约翰·格登爵士两次引用报道。博士后期间,在国际上首次对急性髓系白血病干细胞及造血干祖细胞进行了全基因组甲基化研究,发现了由71个基因组成的白血病干细胞DNA甲基化标记,这个标记与不良预后直接相关,同时揭示了白血病干细胞的两种生物学起源。主要研究成果发表在世界著名学术期刊上,包括Stem CellsNature Communications等一区顶级SCI杂志,参与编撰专著两部,共同主持两项省级重点研发计划,申报国家发明专利五项,多次参加国际学术会议并发表会议摘要二十余篇,同行评审国际学术期刊稿件三十余篇。

科学研究 Scientific Research
戴博实验室专注于人类恶性血液肿瘤,特别是急性髓系白血病(AML)中白血病干细胞的分子/基因组特征和治疗靶向性,同时也关注人的造血干细胞(HSC)和正常造血。我们利用免疫缺陷小鼠异种移植实验来研究和监测异常/正常造血干细胞在体活动,结合生物信息学来确定肿瘤干细胞的关键驱动基因。在世界上首次对人急性髓系白血病(AML)功能性干细胞进行了全基因组甲基化研究,发现了由71个基因组成的功能性肿瘤干细胞DNA甲基化标记。这个标记与不良预后相关。大多数这些基因涉及到表观遗传学改变,与基因突变无关。与此同时,第一个在国际上发现人造血干祖细胞(HSPCs)的甲基化组(methylome)。通过对比肿瘤干细胞与人造血干祖细胞的甲基化组,揭示了人白血病干细胞的两种生物学起源。这项研究率先在国际上发现了直接证据,证明肿瘤干细胞与其子代细胞之间存在表观遗传学差异,并为今后的白血病药物开发提供了靶点。该研究发表于Nature Communications杂志上(6:8489),多次被国际同行报道引用。
