1:钠钾ATP酶及其配体的新生物学功能研究
Na,K-ATPaseisaubiquitousionpumpacrossmembrane,inrecentyears,increasingevidencerevealsthatbesidesion-pumpingactivity,thisiontransporterisalsoactivelyinvolvedinmanypatho-physiologicalregulationviamultiplesignalingpathway.Furthermore,theendogenousNa,K-ATPaseinhibitorsareseparatedandidentifiedfromhumanserumorurine,butthebiologicalfunctionoftheseactivecomponentsremainselusive.OurcurrentprojectistoexplorethenovelbiologicalfunctionofNa,K-ATPaseaswellasitsinhibitorsincancerorinflammation.
2:炎症因子mRNA转录后水平调控机制及其在药物筛选中的应用价值
Thepost-transcriptionalregulationofinflammatorycytokinesplaysacriticalroleinavarietyofimmuneregulation,whichallowsthecellstoelaboratelycontroltheprogressionofimmunereaction.ManystrategiesareadoptedbycellstoregulatecytokinesmRNAturnover,suchasmiRNAorRNAbindingproteins(RBPs).Amongthem,wepayspecialattentiontoRBPssuchasHuR,andtrytouncoverthenovelfunctionoftheseRBPsincontrollingcytokinesmRNAturnover.WearealsointerestedinthespecialintercellularorganellesthatareresponsibleformRNAreprogramming.Basedonthesemechanisms,weexpecttoscreenanti-inflammatoryherbalmedicineextracts,andtoidentifynewchemicalsandnovelmechanisms.
3:小分子抗肿瘤或增敏剂活性物质的寻找与机制研究
Ithaslongbeenrecognizedthatnotallcancercellsaresensitivetoacertainchemotherapeuticdrug,suchasAs2O3.Inviewofthisdeficiency,toincreasetumorcellssensitivitytowardsanti-tumordrugshasbecomeaheattopic.Weshowgreatinterestinlookingforsmallmoleculesthatcansignificantincreasecancercellssensitivitytowardstumor-targetingdrugsfromthelibraryofthenatureproducts,andexpecttoidentifythepromisingdrugtargetforcancertherapy.
4:中药抗肿瘤分子药理学
Theherbalmedicinehasmadegreatcontributiontothediscoveryofnewanti-cancerdrug.Recently,manychemicalsforherbalmedicinehaveshownsignificantanti-tumoreffect,butthemolecularmechanismsremainnotfullyunderstood.Onthisproject,weexpecttoidentifythemechanismsofsomeofthesenaturalproductsbymultiplemolecularaswellascellularapproaches.
