本课题组利用基因芯片、高通量测序等全基因组研究手段,阐明在不同白血病类型中的microRNA的特异表达情况,以及它们与不同种类白血病的发病关系;通过分析基因调控区甲基化水平、基因拷贝数、组蛋白修饰情况等表观遗传学因素,研究microRNA差异表达的机理;同时利用造血干细胞分化培养、RNAi技术、转基因小鼠模型等,在体外和体内系统中分别研究microRNA及其靶基因对于白血病的发生发展、血细胞种系的分化所起到的作用。在药物遗传学(PharmacogeneticsandPharmacogenomics)方面,我们的工作主要利用Genome-WideAssociationStudy,研究个体基因结构与表达差异(如SNP、mRNA、miRNA、CNV等)和多种抗癌药物对个体响应或毒性差异的关联性,为临床医疗和药物试验提供依据。
