研究工作研究方向:淋巴细胞的活化和粘附
T、B淋巴细胞在机体防御中发挥着许多重要的作用:如淋巴细胞迁移到感染或炎症部位,清除致病原;杀伤肿瘤细胞;调节自身免疫性疾病的发生和发展。我们希望利用小鼠细胞和相关疾病模型及样品,通过生化,细胞和分子生物学技术手段,并结合影像(如livingcellimagingunderconfocalortwo-photon)和流式细胞仪等检测方法,研究如下问题:
(1)静息状态(Naïve)下的T、B淋巴细胞如何通过细胞表面受体(如TCR,BCR,趋化因子受体,共刺激分子,整合素等)识别外界信号(如抗原,趋化因子等),并激发细胞内何种信号通路,从而使T、B淋巴细胞发生活化和增殖(见如下TCR和整合素介导的T细胞功能的信号网络图);
(2)整合素是表达在细胞表面的粘附分子,它可以介导细胞粘附到细胞间质或其他细胞表面(如参与T-抗原呈递细胞之间的免疫突触的形成),并调控细胞的增殖、分化、凋亡和迁移等功能。我们将研究整合素如何介导效应T细胞如杀伤性T细胞(CTLs)或B细胞内信号的传导,从而调节T细胞的杀伤功能或抗体的产生;
(3)病理状态下:如在病毒感染中,T、B淋巴细胞如何通过细胞表面受体与病原微生物结合,调节病原微生物在细胞内的复制及在细胞间的传播,从而探索清除病原微生物感染的新策略;或在肿瘤发生中,肿瘤细胞是否及如何调控(manipulate)T细胞(如CTLs或调节性T细胞Tregs),从而逃避T细胞的监控等。
