研究工作Researchinmylabaimstounderstandhowemotionalandsocialbehaviorsareencodedinthebrain,withamainfocusontheneuralcircuitryunderlyingdepressionanddominancehierarchy.Howaredifferentemotionsrepresentedinthebrain?Whereandwhatmolecularchangesoccurinemotionaldisorderssuchasdepression?Howdoesdominancehierarchyarisefrominterplaybetweenactivityofspecificneuralcircuitsandsocialexperienceofanimals?Weareaddressingthesequestionsusingcombinatorialtechniquesincludingimaging,electrophysiology,molecularbiologyandoptogenetic.Wehopetovisualizetheneuralcircuitsthatareactivatedduringthebehaviorofinterest;characterizethefunctionofspecificcircuitsandcellgroupsinbehavior;andestablishthecausalrelationshipbetweentheactivityofneuralcircuitsandbehavioraloutput.Itisourhopethatthesestudieswillprovidenewinsightsintotheneuralcircuitbasisofbehavior,andalsohavetherapeuticimplicationsintreatingemotionaldisorders.Therearecurrentlythreemaindirectionsinthelab:
1.Neuralrepresentationofemotionalvalue
Rewardingandaversiveemotionalstimulievokedistinctpatternsofbehavior.Howdoesthebrainrepresentthesedifferentemotionalvaluesandgeneratecorrespondingbehavioraloutput?Inordertoextractthevaluerepresentationofanemotionalstimulus,wesimultaneouslymaptheneuralensemblesofrewardingandaversiveemotionalstimuli,andcomparetheiractivationpatternsinthesamemousebrain.
2.Molecularandcircuitmechanismofdepression
Thehabenula,anucleiintherewardpathwayandrecentlygarneredalotofattention,encodesthenegativerewardanditshyperactivityhasbeenimplicatedindepression.Weaimtosearchformolecularmechanismsunderlyingthehabenularhyperactivityindepressedanimals.IncollaborationwithDr.JohnYate’slaboratoryattheScrippsInstitute,weperformedahigh-throughputquantitativeproteomicscreentolookformembraneproteinsthatshowalteredexpressionlevelinthehabenulaofcongenitallydepressedrats(fromDr.FritzHenn).Severalproteinsimplicatedinneuronalplasticitywereidentified.Throughthefunctionalcharacterizingofthesecandidatemoleculesinsynapsephysiologyanddepressionbehaviors,wehopetoidentifybiomarkersofdepressionandrevealthekeymolecularmechanismsunderlyingthediseaseetiology.
3.Neuralcircuitmechanismofsocialhierarchy
Dominancehierarchyisafundamentalorganizingmechanismformostsocialanimals.Thesocialeconomicstatushasbeenidentifiedasthesinglestrongestpredictorofhealth.Inarecentwork(Science,2011),weexploredthecircuitryfordominancehierarchybysynapticperturbationinthemedialprefrontalcortex(mPFC),acandidatebrainregionexplicitlyimplicatedinsocialcognition.Weestablishedthatsocialrankinmiceistransitive,relativelystable,andhighlycorrelatedamongdifferentbehavioralmeasures.Furthermore,thehierarchicalstatusofmicecouldbechangedfromdominanttosubordinate,orviceversa,byviralmanipulationofthesynapticstrengthinmPFC.WearecurrentlyusingmolecularandoptogenetictoolstodelineatethemPFCdownstreamcircuitryinvolvedindominancehierarchy.

个人简介胡海岚博士1996年毕业于北京大学生物系,获生物化学和分子生物学专业学士。随后在加州大学伯克利分校师从CoreyGoodman博士,于2002年获得神经生物学方向博士学位。2003-2004年和2004-2008年期间分别在美国弗吉尼亚大学JuliusZhu博士的实验室和冷泉港实验室RobertoMalinow博士的实验室进行博士后工作。于2008年12月加入中国科学院神经科学研究所,担任神经环路与行为可塑性研究组组长。胡海岚博士主要从事情绪与社会行为的分子与神经环路机制研究。
Dr.HailanHuistheHeadoftheLaboratoryofNeuralCircuitandBehavioralPlasticity.Dr.HugraduatedfromBeijingUniversitywithaB.S.degreeinBiochemistry&MolecularBiology(1996).ShepursuedherPh.D.degreeinneuroscienceatUniversityofCaliforniaatBerkeley,whereshewasastudentofProf.CoreyGoodman,workingonrepulsiveaxonguidance.AftercompletingPhD(2002),sheconductedpostdoctoralresearchwithDr.JuliusZhuatUniversityofVirginia(2003-2004),andDr.RobertoMalinowatColdSpringHarborLaboratory(2004-2008),workingonmechanismsregulatingAMPAreceptortraffickinginemotionalbehaviorsanddiseasemodels.Dr.HujoinedIONasaPrincipalInvestigatorinDec2008.Forherpastandcurrentresearches,Dr.HuhasreceivedtheHowardHughesPredoctoralFellowship(1997-2002),theDamonRunyonPostdoctoralFellowship(2003-2006),theChineseHundredTalentPlanAward(2009),andShanghaiPujiangTalentAward(2010).
