Mammalianhibernatorsachievedtheamazinggoalofenergysavingbyactivelysuppressingtheirmetabolicratesundertheextremeenvironmentalconditions.Theyhaveevolvedremarkableabilitiestosustaintherespiratory,cardiovascular,immunological,andneurologicalresponsestometabolicsuppressionduringhibernationthatwouldbelifethreateninginothermammalianspecies.Understandingthemolecularmechanismunderlyingthehibernationphenotypecanprovidethecrucialcluestothedevelopmentofnoveltherapeuticinterventionstohumandiseases.Arcticgroundsquirrel(Spermophilusparryii)providesanexcellentanimalmodeltostudyhibernation.Duringthe6-9monthslonghibernationseason,arcticgroundsquirrelsenterthestateoftorporbyloweringtheircorebodytemperaturestoaslowas–2.9C.However,theyspontaneouslyre-warmtoeuthermicbodytemperatures(36-37C)every10-21daysandmaintainthattemperaturefor15-24hoursbeforeslowlyre-enteringtorpor.Allsmallmammalianhibernatorsexhibitperiodicarousalsfromtorporinspiteoftheexpensiveenergycost.Thefunctionofarousalepisodeshasbeenlargelyunknown.Leadingtheoriesincludethathibernatorsarousetosleepandtoreplenishtheirgeneproducts.Unveilingthebiochemicalprocessesinthetorpor-arousalcyclesinhibernatorswouldhelpustodesignnewtreatmentsforhumanconditionslikestroke,ischemia,andreperfusion.
CollaboratingwiththeresearchersatInstituteofArcticBiologyinUniversityofAlaskaFairbanks,wepreviouslyidentifiedthedifferentialexpressionof625genesinbrownadiposetissue(BAT)withthelarge-scalemousemicroarrayscomparingwintertorpidwithsummeractivearcticgroundsquirrels.Amongthem,thegenesinvolvedinnon-shiveringthermogenesis(NST)weresignificantlyup-regulatedwhereasthoseinvolvedinproteinsynthesisweresignificantlydown-regulated(Yanetal.2006).Followingupthisstudy,weperformedalarge-scalescreeningofdifferentialgeneexpressioninawiderangeoftissuesincludingbrownadiposetissue,liver,heart,hypothalamus,andskeletalmuscleinhibernatingarcticgroundsquirrelscomparingfourstagesintorpor-arousalcyclesandnon-hibernatinganimalsusingbothhigh-throughputilluminabeadarraytechnologyandReal-timePCRassay(Manuscriptinpreparation).Significantseasonaldifferencesofgeneexpressionaredetectedinthegenesinvolvedinglycolysis,fattyacidmetabolism,gluconeogenesis,amino-acidmetabolism,transport,detoxication,cardiaccontractility,circadianrhythms,muscledystrophy,andRNAprotectionetc.invarioustissuescomparingtorpidandarousedanimalswithnon-hibernatinganimals.OurresultscastdoubtonthehypothesisthatmammalianhibernatorsarousetoreplenishtheirgeneproductsonthemRNAlevel.Instead,weobserved,forthefirsttime,complexmodulationofgeneexpressionduringmultiplestagesoftorpor-arousalcycles.Inparticular,mostsignificantdifferencesofgeneexpressionduringtorpor-arousalcyclesarefoundduringthetransitionfromlatetorportoearlyarousal.Duringthistransition,themRNAlevelofagroupofmetabolicgenessignificantlydropsperhapsduetotheexhaustionofmRNAtranscriptsduringtheenergeticearlyarousalphase,whereasthemRNAlevelforthegenesrelatedtocellgrowthandproliferationrisessharplywhichmayreflecttheresumptionofcellcycleduringarousalthathasbeenstalledduringtorpor.
