蛋白质泛素化修饰与癌症发生机制
泛素蛋白酶体系统在DNA复制、DNA修复、转录、蛋白合成、细胞分化、凋亡等生命活动中具有重要调节作用。蛋白泛素化过程中包含E1泛素化激活酶,E2泛素化连接酶,E3泛素化蛋白连接酶的参与。泛素蛋白自身含有76个氨基酸残基,分子量为8.5kDa.其重要特点是其含有7个Lysine残基。许多RINGFINGER蛋白功能上作为E3连接酶,诱导底物泛素化和蛋白水解酶体依赖的降解。目前我们主要集中下面三个方面的研究:1)PPARγE3ligase功能机制;2)PPARs(PPARα、PPARγ、PPARδ)调节肿瘤发生分子机制;3)新的PPARs配体合成、筛选及其临床运用。
Theubiquitin-proteasomesystemfunctionsinawidevarietyofcellularprocesses,includingDNAreplication,DNArepair,transcription,proteinsynthesis,celldifferentiationandapoptosis.UbiquitinationiscarriedoutbyanenzymaticcascadecomposedoftheE1ubiquitin-activatingenzyme,theE2ubiquitin-conjugatingenzymeandtheE3ubiquitin-proteinligase.LotsofstudiesshowthatLys-63-linkedchainsisassociatedwithsignaltransduction,membrane-proteintrafficking,endocytosisandDNArepair,incontrast,K6,K11,K27,K29,K33,K48linkedubiquitinchainstargetproteinsforproteasome-mediateddegradation.Atpresent,wefocusthreeareasasfollows:
1)ThefunctionalmechanismofPPARγE3ubiquitinligase.
2)ThemolecularmechanismofPPARs(PPARα,PPARγ,PPARδ)regulatestumorigenesis.
3)synthesisandscannovelPPARsligandstotreatdiseases.
