马慧敏,副研究员,博士,从事环境污染物的早期健康效应研究,研究了大气细颗粒物(包括PM2.5和PM0.1等),环境中的重金属和有机物(苯,三氯生等)等对人体健康的影响,利用体内体外实验方法,筛选大气颗粒物中的有效毒性组分,筛选重金属对人体健康影响的安全阈值,筛选环境有机污染物等对人体的特异性生物标志物,阐述对人体的毒作用模式。
针对环境中重金属的污染问题,对广州地区一个重金属污染区的适龄儿童进行智力测试,结合儿童的血液和尿液中的四种重金属的水平数据,分析出四种重金属之间有显著的联合效应,其中血铅仍然是诱导儿童智力降低的主要因素,结合体外实验,筛选出2个印记基因DNA甲基化生物标志物。结果发现血铅在低于国家检测线100 μg/L 的情况下(约57 μg/L以下),也会和智力有显著的负相关性,提示了国家政府部门需要对血铅的相关安全标准再重新进行审视,结论发表在ES&T和EP杂志上。
分析PM2.5样品各组分中的有效毒性物质,初步结果表明大气颗粒物中类腐殖酸类化合物(Huminc-like Substances, HULIS)可能是PM2.5中诱导炎症和凋亡的主要毒性组分,并随后开展了人群样品的HULIS毒性鉴定。利用细胞模型检测毒性,傅里叶变换离子回旋共振质谱仪分析化合物组成信息,加速器质谱等分析不同污染物的来源信息,核磁共振等技术探索有机物结构信息,制备性液相色谱分离不同化合物组分等。揭示了大气颗粒物中的HULIS组分可能是颗粒物水溶性组分中的诱导炎症效应和凋亡的主要组分之一,其中含氧基团,芳香环和双键结构等对炎症和凋亡的贡献较大,这些化合物可能与玉米秸秆等生物质燃烧、交通源排放和燃煤排放有关。开展了人群定组研究(panel study),采集了定组人群的样品,分析大气PM2.5/PM0.1暴露对人体心肺等功能损伤,颗粒物各组分对健康的危害及其贡献大小,各组分的毒作用机制,研究结果发表在ES&T和EP等杂志上,并选为当期的内封面。
在国内首次建立HPLC-MS/MS全基因组DNA甲基化和MALDI-TOF-MS/MS分析组蛋白修饰的检测方法,分析了环境中普遍存在的三氯生和苯及其代谢物低浓度暴露诱导的毒性效应,并筛选出了潜在的分子标志物。关于三氯生的文章,发表在毒理学专业的期刊Toxicology in vitro 和Chemosphere上,被英国知名的文献推荐平台F1000prime推荐为“非常好”的论文,并评价了论文结果对三氯生肝毒性的贡献。
