  干细胞是机体组织器官再生的源泉,其自我更新与分化之间的动态平衡对于组织器官的生长发育和机能维持具有重要的作用。在人类的一生中,干细胞经历了蓬勃发育、维持和衰退的过程。这一过程受到严格的时空调控,而干细胞命运调控机制的失衡与人类疾病的发生息息相关。在衰老过程中,干细胞逐渐耗竭,是导致衰老和衰老相关疾病的发生的重要原因。
  课题组主要围绕干细胞的命运决定和稳态维持,通过结合基因编辑技术、多谱系的干细胞分化技术、多层次组学手段、新型的动物模型等,系统研究再生与衰老的分子机制,特别是表观遗传因素在机体退化和病变中发生的作用。 取得的主要研究成果包括:(1)在衰老的遗传和表观遗传学基础方面,结合基因编辑、重编程和多能干细胞定向分化技术,建立儿童早衰症和成年早衰症等人类干细胞疾病模型,揭示了细胞核结构和表观遗传改变是人类早衰症和生理性衰老共同的驱动力(Science, 2015; Cell, 2016; Protein and Cell, 2018);(2)建立了简便可扩大的人类干细胞平台,用于延缓衰老化合物的筛选评估和全基因组范围内关键的衰老调控基因的筛查。基于这一体系,目前已经发现如奥替普拉等小分子可以有效的延缓早衰和野生型人类干细胞衰老,为治疗早衰症和延缓自然衰老提供了思路和依据 (Cell, 2016; Protein and Cell, 2016)。3)利用长寿基因SIRT6敲除的基因工程猴,研究灵长类动物不同类型器官、组织、细胞中受SIRT6调节的分子信号网络 (Nature, 2018)。
