1.量子点超敏荧光免疫传感
荧光免疫层析技术是当前应用于即时诊断(POCT)领域的重要平台,而开发高性能荧光标记探针则是提升免疫层析技术检测性能的关键所在。为解决荧光标记探针发光单元强度弱、荧光团负载量低等缺陷,我们发展了基于金属-配基亲和驱动组装策略的新型纳米结构调控方法,将高色纯度以及高亮度的油溶性量子点紧密、均匀封装于胶体二氧化硅微球基质。基于该探针的荧光免疫层析检测卡可与便携式荧光扫描仪配合使用,在10~15min内可直接检出血液、尿液等样本中的特定生物标志物(如炎症标志物、肿瘤标志物、心肌标志物等),具有高灵敏及特异性优势。利用类似原理,该技术同样应用于毒品检测(如冰毒)、传染病诊断以及食品安全检测(如毒素)等领域的超敏即时诊断。
2.纳米磁珠生物纯化技术与产业化应用
我们基于功能单元的控制合成及其组装过程优化的多维材料设计理念,结合纳米模板与亲和驱动策略,实现了对磁性及光学等纳米原件的可控组装及功能集成与协同。力求解决当前磁分离纯化过程中,分离单元磁响应性弱、多分散、液相分散性差等制约体外诊断与生物纯化性能的关键问题。成功制备了单分散、快速磁响应、低沉降、重悬性好(超顺磁)的系列纳米生物磁珠。目前已成功应用的领域包括:在特定条件下吸附/脱附核酸,用于全血、组织、细菌、病毒、植物等DNA/RNA的提取纯化;基于自动化磁分离平台与分子诊断技术(PCR)的血液病毒筛查(乙肝、丙肝、艾滋多重检测)、刑侦司法用痕量样本(指纹、毛发、皮肤等)DNA提取及检测;用于医院及第三方检验机构磁珠法化学发光平台对血液中生物标志物的高通量自动化检测,并与上市公司宁波美康生物开展合作共同推进化学发光用免疫磁珠国产化。
