Ourgroupcurrentresearchinterestsarethedevelopmentofnewsyntheticmethodologies,strategiesandtransition-metalcatalyzedtandemprocessesfortherapidconstructionofbiologicallyactiveheterocycles.
(1)Transition-metalCatalyzedDecarboxylativeCross-couplingReaction
Transition-metalcatalyzeddecarboxylativecross-couplingreactionsusingsimplecarboxylicacidsascouplingpartnershaveemergedasanewtypeofC-Cbondformationreaction.Inthesereactions,usingreadilyavailableandstablecarboxylicacidsinsteadoforganometallicreagentsenabledthedecarboxylativecross-couplingreactionstoproceedwithgoodselectivitiesandtoleranceoffunctionalgroups(Scheme1).

Wehavedevelopedaseriesoftransition-metalcatalyzeddecarboxylativeacylationandacyloxylationofdifferentreactantswitha-oxocarboxylicacidviadirectunactivatedC–Hbondfunctionalizationundermildconditions(Scheme2,a-c).
Anefficientcatalyticdecarboxylativeacylationofunactivatedsp2(alkenyl)C-Hbondshasbeendeveloped.Varioussubstituteda-oxocarboxylicacidswithdifferentelectronicpropertiesreactundermildconditionstoaffordadiverserangeofb-acylenamideproductsingoodyields.Thereactionisproposedtoproceedviaacyclicvinylpalladiumintermediate,facilitatingthedecarboxylativedehydrogenativeprocesswithenamidecouplingpartners(Scheme2,a).
Anovelmetal-freedecarboxylativeacyloxylationofsp3C-Hbondinformamidesandethershasbeenexplored.AvarietyofN-acyloxymethylamidesanda-acyloxyetherscouldbeeasilysynthesizedbythismethod.Preliminarymechanisticstudieshaveshownthatthereactionproceededviaaradicalprocess(Scheme2,b).
Amildandefficientsilver-catalyzedacylarylationofactivatedalkeneswitheasilyavailablea-oxocarboxylicacidshasbeendeveloped.Thereactionsproviderapidaccesstoavarietyoffunctionalizedoxindolesviaatandemdecarboxylativeradicalcyclizationstrategy.Thistransformationproceedswellundermildreactionconditionsandexhibitsexcellentfunctionalgrouptolerance,affordingthedesiredoxindolesingoodtoexcellentyields(Scheme2,c).Wehavealsodescribedseveraloxidativecouplingreactionsusinga-oxocarboxylicacidsasreactants(Scheme2,dande).

(2)SynthesisofHeterocycles
Heterocyclesrepresentalargergroupoforganiccompoundsandplayasignificantroleinlifeprocessedandinallaspectsofpureandappliedchemistry.Nitrogen-containingheterocyclesareancomponentofnotonlysomeofthemostinterestingandbiologicallyimportantnaturalproducts,suchasalkaloids,carbohydrates,nucleicacids,andantibioticsbutincludenumerouspotentpharmaceuticaldrugsandalargeamountofsyntheticorganiccompounds(Scheme3).

Oxindoles,spirooxindolesanddihydroquinolinones,importantandrepresentativenitrogenheterocyclesareofgreatinteresttoorganicsynthesisduetotheirsignificantbiologicalactivitiesandtheirwideapplicationsinorganicsynthesis.Inthepastfewmonths,aseriesofpracticalprocessesforthesynthesisofthesemotifshavebeendevelopedbyourgroup(Scheme4).
