林厚文教授是国家****、****、上海市优秀学术带头人。长期聚焦海洋共生生物海绵及其共附生微生物活性分子的化学与生物学研究,通过整合“微生物生态-天然药物化学-分子生物学-合成生物学”研究平台,借助高分辨多级质谱-分子网络数据库分析(HRMSn-MNDA)等微量新颖天然产物的快速精准发现技术,解析了1500 余种复杂天然分子的结构,新化合物 528 个,国际热点化合物20余个,HipplachninA 被美国化学会评选为“海洋天然产物复兴”代表之一;构建临床肿瘤干细胞筛选平台,获得抗肺癌干细胞耐药、靶向 Chk1 新型小分子抑制剂等 10 个海洋抗肿瘤先导化合物,阐明了其作用机制;解析了重要药源分子抗肿瘤缩酚酞NAT的生物合成机制并构建其高效生物合成体系,并获得活性改良的新结构。
