摘要/Abstract
摘要: 目的 ·观察抗性糊精对 db/db小鼠胰岛素抵抗状态的影响,并初步探索潜在的分子机制。方法 ·取 8周龄 db/db小鼠 10只,随机分为对照组和抗性糊精组,分别予蒸馏水和抗性糊精灌胃 9周。每周检测小鼠空腹血糖( fasting blood glucose,FBG)及体质量,并于第 8周行腹腔注射葡萄糖耐量试验( intraperitoneal glucose tolerance test,IPGTT),于第 9周行胰岛素耐量试验( insulin tolerance test,ITT),处死小鼠后测定血清空腹胰岛素( fasting serum insulin,FINS)、三酰甘油( triacylglycerol,TAG)、总胆固醇(total cholesterol,TC)、低密度脂蛋白胆固醇( lower density lipoprotein-cholesterol,LDL-Ch)、高密度脂蛋白胆固醇( high density lipoprotein-cholesterol,HDL-Ch)的水平并计算稳态模型胰岛素抵抗( homeostasis model assessment insulin resistance,HOMA-IR)指数,实时荧光定量 PCR检测肝脏组织胰岛素信号转导通路 Ir、Irs-1、Pi3k、Akt、Glut-2 mRNA相对表达量, Western blotting检测肝脏 IRS-1、p-AKT、GLUT-2蛋白的表达。结果 ·与对照组相比,经抗性糊精灌胃处理 9周后,抗性糊精组小鼠体质量降低,但差异无统计学意义( P0.384);FINS有所下降但差异无统计学意义, HOMA-IR指数显著下降( P0.032);胰岛素抵抗得到显著改善(均 P<0.05);血生化指标 TAG、TC、LDL-Ch、HDL-Ch均降低,但仅 LDL-Ch的差异具有统计学意义(P0.034);Ir、Irs-1、Akt、Glut-2 mRNA表达显著增加 (均 P<0.05);IRS-1、p-AKT、GLUT-2蛋白表达量增加,其中 IRS-1和 GLUT-2表达量的升高具有统计学意义(P0.026,P0.039)。结论 ·抗性糊精可能通过增强胰岛素信号转导改善 db/db小鼠的胰岛素抵抗。

Abstract:
Objective · To investigate the effect of resistant dextrin on insulin resistance in db/db mice and preliminarily explore potential molecular mechanisms. Methods · Ten 8-week-old db/db mice were randomly assigned into two groups, i.e., control group and resistant dextrin group, which were administered with distilled water and resistant dextringavage for 9 weeks, respectively. Fasting blood glucose (FBG) and body weight were measured every week. Intraperitoneal glucose tolerance test (IPGTT) and insulin tolerance test (ITT) were performed separately at the 8th and 9th week. Fasting serum insulin (FINS) was measured, homeostasis model assessment insulin resistance (HOMA-IR) index was calculated, and triacylglycerol (TAG), total cholesterol (TC), low density lipoprotein-cholesterol (LDL-Ch), high density lipoprotein-cholesterol (HDL-Ch) were detected after the mice were sacrificed. The relative of Ir, Irs-1, Pi3k, Akt, and Glut-2 in insulin signaling pathways in the liver was determinedreal-time quantitative PCR. The of IRS-1, p-AKT, GLUT-2 in the liver were determinedWestern blotting. Results · After 9 weeks of treatment with resistant dextrin, the body weight of the resistant dextrin group decreased, but the difference was not statistically significant (P0.384); FINS decreased, and HOMA-IR index significantly decreased (P0.032); insulin resistance was significantly improved (all P<0.05); the blood biochemical parameters, including TAG, TC, LDL-Ch, and HDL-Ch were lower, but only LDL-Ch was statistically different (P0.034); the of Ir, Irs-1, Akt, and Glut-2 mRNA in the liver significantly increased (all P<0.05); the of IRS-1 and GLUT-2 proteins also significantly increased (P0.026, P0.039). Conclusion · Resistant dextrin may improve insulin resistance in db/db miceenhancing insulin signaling pathway.
