厌氧氨氧化菌是一种化能自养菌,它具有独特的生理生化特性及生物脱氮机理.对厌氧氨氧化菌进行实验室扩大培养,对其主要代谢酶联氨氧化酶性质进行研究,对联氨氧化酶进行初步纯化,并探究醌类化合物对联氨氧化酶活性的影响.最终得出联氨氧化酶活性最适温度为35 ℃,最适pH为7.5.酶纯化结果表明超滤相对分子质量在50~100 kDa.投加辅酶Q对提高酶活性效果最好,2-羟基-1,4萘醌对酶活性提高效果稍差,蒽醌-2-磺酸钠盐对酶活性基本无影响.
英文摘要:
AnAOB is a kind of chemoautotrophic bacteria, which has unique physiological and biochemical characteristics and biological denitrification mechanism. The laboratory expanding culture of AnAOB is conducted, the property of its main metabolic enzyme hydrazine oxidizing enzyme (HZO) is studied, the preliminary purification of HZO is carried out, and the effect of quinones on the activity of HZO is discussed. It is found that the optimum enzyme activity temperature of HZO is 35 ℃, the optimum pH is 7.5. Enzyme purification result shows that the relative molecular mass of ultrafiltration is 50-100 kDa. Coenzyme Q has the best effect on improving enzyme activity, 2-hydroxy-1,4 naphthoquinone has a less effect on enzyme activity improvement, and anthraquinone-2-sulfonate has no effect on enzyme activity.
