目的探讨膜联蛋白A7(ANXA7)及血管内皮生长因子(VEGF)在乳腺癌组织中的表达及其临床意义。方法应用免疫组织化学方法分别检测94例乳腺癌及其癌旁组织中ANXA7蛋白及VEGF蛋白表达情况,分析ANXA7、VEGF蛋白表达与患者年龄、肿瘤大小、病理分级、TNM分期、淋巴结转移、人类表皮生长因子受体2(HER-2)、雌激素受体(ER)、孕激素受体(PR)表达情况、复发、远处转移等临床病理参数的关系。所有患者随访3年,分析两组不同ANXA7、VEGF蛋白表达患者乳腺癌复发率、转移率。结果乳腺癌组织ANXA7、VEGF蛋白阳性率显著高于癌旁组织(P < 0.05)。随TNM分期升高,乳腺癌组织中ANXA7、VEGF蛋白阳性率升高,病理分级Ⅲ级、HER-2阳性、淋巴结转移乳腺癌组织ANXA7、VEGF蛋白阳性率显著高于病理分级Ⅰ~Ⅱ级、HER-2阴性、无淋巴结转移乳腺癌组织(P < 0.05)。ANXA7阴性组复发率、转移率显著低于ANXA7阳性组(P < 0.05);VEGF阴性组与VEGF阳性组复发率、转移率比较无统计学差异(P>0.05)。ANXA7蛋白阳性乳腺癌组织中VEGF蛋白阳性率显著高于ANXA7蛋白阴性乳腺癌组织(P < 0.05),Spearman相关分析显示,乳腺癌组织中ANXA7蛋白表达与VEGF蛋白表达呈正相关(P < 0.05)。结论乳腺癌组织中存在ANXA7、VEGF蛋白异常高表达,两者在乳腺癌发生、发展中可能起协同作用,并与乳腺癌的病理分级、TNM分期及淋巴结转移有一定关系。

ObjectiveTo investigate the clinical significance of annexin A7 (ANXA7) and vascular endothelial growth factor (VEGF) expression in breast cancer.MethodsThe expression of ANXA7 and VEGF in 94 cases of breast cancer and adjacent tissues were detected by immunohistochemistry, The relationship between ANXA7, VEGF protein expression and clinicopathological parameters, such as age, tumor size, pathological grade, TNM stage, lymph node metastasis, expression of human epidermal growth factor receptor 2 (HER-2), estrogen receptor (ER), progesterone receptor (PR), recurrence and distant metastasis were analyzed. All patients were followed up for 3 years to analyze the recurrence rate and metastasis rate of breast cancer in two groups with different expression of ANXA7 and VEGF.ResultsThe positive expression frequency of ANXA7 and VEGF in breast cancer tissues were significantly higher than those in adjacent tissues (P < 0.05). The positive rate of ANXA7 and VEGF in breast cancer tissue increased with pathological grading. The positive expression frequency of ANXA7 and VEGF in pathological grading, Ⅲ HER-2 positive and lymph node metastasis breast cancer tissues were significantly higher than those in pathological grading Ⅰ-Ⅱ, HER-2 negative and non lymph node metastasis breast cancer tissues (P < 0.05). ANXA7 expression was positively correlated with VEGF expression in breast cancer tissues (P < 0.05). The rate of recurrence and metastasis in ANXA7 negative group were significantly lower than that in ANXA7 positive group (P < 0.05), whereas there was no significant difference between VEGF negative group and VEGF positive group (P>0.05).ConclusionThere is abnormal high expression of ANXA7 and VEGF protein in breast cancer tissue, which could play a role in the occurrence and development of breast cancer.
