定喘汤治疗呼吸道合胞病毒感染的毛细支气管炎的代谢组学研究

Abstract: Objective Based on metabonomics, this study analyzes the therapeutic effect of Dingchuan decoction on bronchiolitis caused by respiratory syncytial virus (RSV). Methods Thirty children with RSV bronchiolitis treated in our hospital between September, 2016, and February, 2017, were selected as research subjects, and 38 healthy children examined in our hospital during the same period were selected as the control group. Metabonomic analysis was performed using LC-MS/MS to compare the metabolites between the two groups. All children in the capillary group were treated with Dingchuan decoction, and the therapeutic effect of the Dingchuan decoction was investigated using metabonomics to elucidate its mechanism of action. Results Metabonomic analysis revealed significant metabolic differences in children with RSV bronchiolitis compared to healthy children, primarily involving three lysophosphatidylcholine (LPC) mole-cules:LPC 18:2 sn-1, LPC 18:0 sn-1, and LPC 18:0 sn-2. Additionally, phenylalanine (Phe), and three phosphatidylcholines (PC 34:3, PC 34:2, and PC 34:1) showed significant metabolic changes. In children with RSV bronchiolitis treated with the Dingchuan decoction, notable metabolic changes were primarily observed in three LPC molecules:LPC18:0 sn-1, LPC 18:0 sn-2, and LPC 16:0 sn-1. Additionally, Phe, PC 34:2, SM34:1, and norvalerolac (Delta-valerolactam) displayed significant differences in metabolic profiles before and after treatment. Notably, the most pronounced changes were observed in LPC 18:0 sn-1 and LPC 18:0 sn-2. Conclusion Metabolomics analysis could be used to distinguish the different metabolic patterns in children with RSV bronchiolitis before and after treatment with Dingchuan decoction. Moreover, the most significant difference in lipid metabolites could be considered as an important mechanism of the Dingchuan decoction exerting its therapeutic effect.
