组织芯片制备仪联合STR检测在葡萄胎病理诊断中的应用

摘要: 目的 探讨应用组织芯片制备仪精准分离绒毛组织和蜕膜组织联合短串联重复序列(STR)分析在葡萄胎病理诊断中的应用价值。方法 收集中国医科大学附属盛京医院2017年3月至2022年9月可疑葡萄胎的组织标本90例。标本常规固定、脱水及石蜡包埋。分别应用组织芯片制备仪、传统方法分离标本中绒毛组织和蜕膜组织,应用检测试剂盒进行STR检测。结果 应用组织芯片制备仪STR检测成功率为95.6%,采用传统方法STR检测成功率为60.0%。成功进行STR检测86例,包括葡萄胎67例和非葡萄胎19例。结论 应用组织芯片制备仪精准分离绒毛组织联合STR分析技术提高了STR检测的成功率,在葡萄胎病理诊断中具有重要意义。

Abstract: Objective To investigate the applicability of short tandem repeat (STR) analysis in the precise separation of villus and decidua tissue using tissue chip preparator in the pathological diagnosis of hydatidiform mole (HM). Methods Suspected HM tissue specimens (n=86) were collected from Shengjing Hospital of China Medical University between March 2017 and September 2022. The specimens were fixed,dehydrated,and embedded in paraffin. The villus and decidua tissue were separated using a tissue chip preparation device and a traditional method,and STR were detected using a testing kit. Results The STR detection rate using the tissue chip preparator and traditional method was 95.6% and 60%,respectively. In this study,86 cases of STR were successfully detected,including 67 HM and 19 and non-mole cases. Conclusion The combined application of the tissue chip preparator to accurately separate villi and STR analysis technology can improve STR detection rate,which has important significance in the pathological diagnosis of HM.
