系统性红斑狼疮患者外周血中T细胞α7nAChR mRNA的表达及其临床意义

Abstract: Objective To investigate the expression and clinical significance of α7 nicotinic acetylcholine receptor(α7nAChR)mRNA in peripheral blood T cells of patients with systemic lupus erythematosus(SLE). Methods We recruited 16 patients with SLE(the SLE group)who received treatment at the Department of Rheumatology and Immunology,The First Hospital of China Medical University. Additionally,we enrolled 16 healthy individuals matched for sex and age as the control group. Blood samples were collected prior to treatment. Various tests were conducted,including peripheral blood cell counting for white blood cells and lymphocytes,immunoglobulin assays, complement assays,C-reactive protein(CRP)tests,erythrocyte sedimentation tests,cytokine expression tests,and T cell subset tests. The α7nAChR mRNA expression in CD4+ and CD8+ T cells was analyzed using real time PCR in the SLE group. Pearson’s correlation or Spearman’s correlation analysis was used to explore the correlation between the clinical indicators and their corresponding α7nAChR mRNA expression in patients with SLE. Results Compared with the control group(1.02±0.49,1.83±0.46),expression of α7nAChR mRNA in CD4+ and CD8+ T cells was significantly lower in the SLE group(0.50±0.27,P=0.005;0.83±0.58,P=0.001). Notably,in SLE patients,the level of α7nAChR mRNA in peripheral CD4+ T cells exhibited a significant negative correlation with serum IgG (r=-0.637,P=0.035),CRP(r=-0.611,P=0.034),and systemic lupus erythematosus disease activity index(SLEDAI) (r=-0.605, P=0.030). Conclusion The expression of α7nAChR mRNA is low in the peripheral blood T cells of patients with SLE.α7nAChR mRNA expression was negatively correlated with SLEDAI scores,serum IgG levels,and CRP levels. These results suggest that α7nAChR may be involved in the pathogenesis and progression of SLE and could serve as a novel clinical indicator for assessing SLE disease activity.
