摘要: 目的 建立铜死亡相关配对长链非编码RNA(lncRNA)肺腺癌预后模型,探讨其预后和临床意义。方法 由癌症基因组图谱(TCGA)数据库获取肺腺癌相关lncRNA,与铜死亡相关基因进行相关性和差异性分析,得到铜死亡相关差异lncRNA。使用多种算法,筛选出预后相关配对lncRNA来构建模型。对高风险组和低风险组进行生存分析、基因功能富集分析、基因突变分析和药物反应预测。结果 得到3 647个铜死亡相关配对lncRNA,经过严格筛选,最终整合6个预后相关配对lncRNA建立预后模型。生存分析显示,高风险组较低风险组预后更差(P<0.000 1)。单因素和多因素Cox回归分析均提示,风险评分为独立预后因素。高风险组中,多数化疗药物(如顺铂、紫杉醇等)表现出更低的半数抑制浓度。结论 本研究建立的铜死亡相关配对lncRNA肺腺癌预后模型,有助于预测肺腺癌患者的预后、生物学特征和潜在治疗药物。

Abstract: Objective To establish a prognostic signature of cuproptosis-related long noncoding RNA(lncRNA)pairs in patients with lung adenocarcinoma(LUAD)and explore the signature’s usefulness for LUAD prognosis as well as its clinical significance. Methods Samples of lncRNA associated with LUAD were obtained from the Cancer Genome Atlas database. Differential correlation analysis with cuproptosis-related genes was carried out to confirm the differentially expressed cuproptosis-related lncRNA pairs(DEcrlncRNA pairs). The signature was constructed by using a variety of algorithms to obtain DEcrlncRNA pairs that were associated with prognosis. Survival analysis,gene enrichment analysis,gene mutation analysis,and drug response prediction were also carried out for highand low-risk groups. Results A total of 3 647 DEcrlncRNA pairs were obtained to integrate six prognosis-related DEcrlncRNA pairs into a prognostic signature. Survival analysis indicated that prognosis for the low-risk group was worse than for the high-risk group(P<0.000 1). Both univariate and multivariate Cox regression analyses suggested that risk scores were independent prognostic factors. The half maximal inhibitory concentration for the majority of chemotherapy drugs(e.g. cisplatin and paclitaxel)was lower in the high-risk group. Conclusion The prognostic signature of cuproptosis-related lncRNA pair signatures of patitents with LUAD established in this study may help determine the biological characteristics of LUAD,as well as identify prognoses and potential therapeutic drugs for patients with LUAD.
