miR-183-5p和NDUFA4L2在肾透明细胞癌中的表达及相关性

Abstract: Objective To detect the characteristics of the expressions of microRNA-183-5p (miR-183-5p) and NDUFA4L2 in renal clear cell carcinoma and analyze their correlation and effect on postoperative survival time. Methods A total of 57 patients with renal clear cell carcinoma who underwent surgery were included, and renal cancer and adjacent normal renal tissues were collected. The expressions of miR-183-5p and NDUFA4L2 were detected by real-time fluorescence quantitative PCR (qRT-PCR) and immunohistochemistry, respectively. The expressions of miR-183-5p and NDUFA4L2 mRNA in human renal clear cell carcinoma cell line 786-0 and human renal pro-ximal tubular epithelial cell line HK-2 were detected by qRT-PCR. Results The expression level of miR-183-5p was significantly lower and the positive expression rate of NDUFA4L2 was significantly higher in renal cancer tissues than that of adjacent normal tissues (P < 0.05). The expression level of miR-183-5p and the positive expression rate of NDUFA4L2 were significantly different in various WHO/ISUP grades, renal sinus invasion, and maximum tumor diameters (P < 0.05). The expression level of miR-183-5p and NDUFA4L2 score were correlated with postoperative survival time. A negative correlation was found between expression level of miR-183-5p and NDUFA4L2 score. The expression of miR-183-5p was significantly lower and the expression of NDUFA4L2 mRNA was significantly higher in the 786-0 cell line than that of the HK-2 cell line (P < 0.05). Conclusion miR-183-5p and NDUFA4L2 are abnormally expressed and nega-tively correlated in renal clear cell carcinoma, which can promote lesion formation and progression. The expressions of miR-183-5p and NDUFA4L2 may be related to the prognosis.
