Toll样受体4参与卵巢癌细胞增殖与凋亡的调控

Abstract: Objective To investigate the effects of abnormal expression of Toll-like receptor 4 (TLR4) on the proliferation and apoptosis of ovarian cancer cells. Methods TLR4 expression in the ovarian cancer cells and surrounding normal tissues was detected by immunohistochemistry. siRNA targeting TLR4 gene was constructed and transfected into human ovarian cancer cell line SKOV3. TLR4 mRNA and protein expression levels were detected by real-time quantitative polymerase chain reaction,and Western blotting,respectively. Cell counting kit-8 method assay was used to measure viability of SKOV3 cells after transfection with TLR4 siRNA for 6-24 h,and flow cyto-metry was used to detect the apoptosis of SKOV3 cells after transfection for 24 h. Results TLR4 was expressed in both ovarian cancer cells and adjacent normal tissues;however the expression level of TLR4 in the ovarian cancer tissues was significantly higher than that in the adjacent normal tissues (P<0.05). After siRNA transfection,the expression levels of TLR4 mRNA and protein in the SKOV3 cells were significantly decreased (P<0.05). After 24 h siRNA transfection,SKOV3 cell viability decreased,whereas the apoptosis rate increased (P<0.05). Conclusion TLR4 is involved in the regulation of ovarian cancer cell proliferation and apoptosis,and interference with TLR4 can inhibit SKOV3 cell proliferation and induce apoptosis.
