STEAP3在脑缺血再灌注损伤沙鼠中的表达及意义

摘要: 目的 探讨前列腺六跨膜上皮抗原3 (STEAP3)在脑缺血再灌注(I/R)损伤沙鼠中的表达及意义。方法 将48只沙鼠随机分为假手术组(Sham组)、I/R模型组(I/R组)、I/R模型+STEAP3 siRNA组(siRNA组)和I/R模型+scramble siRNA组(Control组),每组12只。进行沙鼠神经功能评分。Nissl染色观察海马CA1区神经元。Fe2+试剂盒检测海马组织Fe2+水平。Western blotting检测海马组织中STEAP3、H2AX、4-HNE、GPX4蛋白的表达。应用丙二醛(MDA)试剂盒检测海马组织中MDA含量。结果 I/R组沙鼠神经功能学评分、STEAP3、H2AX、4-HNE蛋白及MDA和Fe2+水平明显高于Sham组(P<0.05),而神经元数量和GPX4蛋白表达明显低于Sham组(P<0.01);与I/R组比较,siRNA组沙鼠神经功能学评分、STEAP3、H2AX、4-HNE蛋白及MDA和Fe2+水平明显降低(P<0.05),神经元数量和GPX4蛋白表达明显增多(P<0.05)。结论 STEAP3在沙鼠脑I/R损伤中高表达,通过介导铁死亡影响脑I/R损伤的发生和发展,抑制其表达对脑I/R损伤具有保护作用。

Abstract: Objective To explore the expression of six-transmembrane epithelial antigen of prostate 3 (STEAP3) and its significance in gerbils with cerebral ischemia-reperfusion (I/R) injury. Methods Forty-eight gerbils were randomly divided into four groups:a sham group, an I/R model group (I/R group), an I/R model+STEAP3 siRNA group (siRNA group), and an I/R model+scramble siRNA group (control group), with n=12 per group. Neurological function was scored. Nissl staining was performed to observe the neurons in the hippocampus. The expression of STEAP3, H2AX, 4-HNE, and GPX4 in the hippocampus was measured using Western blotting. The levels of malondialdehyde (MDA) and Fe2+ were assessed. Results The neurological function scores, the expression of STEAP3, H2AX, and 4-HNE, and the levels of MDA and Fe2+ were significantly higher in the I/R group (P<0.05) than in the sham group, and the expression of GPX4 and the number of normal neurons were significantly lower (P<0.01). The neurobehavioral scores, the expression of STEAP3, H2AX, and 4-HNE, and the levels of MDA and Fe2+ were significantly lower in the siRNA group (P<0.05) than in the I/R group, and the expression of GPX4 and the number of normal neurons were significantly higher (P<0.05). Conclusion Increased expression of STEAP3 was found in gerbils with cerebral I/R injury, which may affect the occurrence and development of cerebral I/R injury by mediating ferroptosis. Knockout of STEAP3 can protect the brain from I/R injury.
