Abstract: Objective To evaluate the expression level of the activating transcription factor 2(ATF2) in pancreatic cancer (PC) cells,and to explore the relationship between ATF2 and PC cells proliferation and apoptosis.Methods Real-time PCR and Western blotting techniques were used to detect the expression of ATF2 messenger RNA (mRNA) and protein in 14 samples of PC tissues and adjacent normal tissues (ANT).ATF2 short hairpin RNA (shRNA) was constructed to transfect the PC cell lines,and cell proliferation and apoptosis were evaluated in the PC cells after the transfection.Results The expression levels of ATF2 mRNA and protein in the PC tissues were higher than in the ANT (P < 0.05),and the expression level of phosphorylated ATF2(p-ATF2) protein was also higher in the PC tissues (P < 0.01).Compared with the control group,the proliferation of PC cells transfected with ATF2 shRNA was inhibited,while apoptosis was increased (P < 0.01).Conclusion The expression level of ATF2 is increased in PC cells.Transfection of PC cells with ATF2 shRNA can significantly inhibit cell proliferation and increase cell apoptosis.
