幽门螺杆菌阳性胃癌中vacA基因型的分布及其与MEX3A表达的关系

Abstract: Objective To investigate the distribution of vaculating toxin geng A(vacA) dominant genotype and its effect on MEX3A protein expression in Helicobacter pylori(H.pylori) -positive gastric cancer tissues. Methods Totally 98 patients who underwent surgical excision of H.pylori-positive gastric cancer in the Central Hospital of Shenyang Medical College from 2011 to 2014 were enrolled in this study. DNA was extracted from the gastric cancer tissues using the GT pureTM FFPE tissue DNA Extraction Kit. vacA genotypes were determined through polymerase chain reaction, and MEX3A protein expression in the cancer tissues was detected using immunohistochemistry. Results The distribution of vacA s1,vacA m1, and vacA s1m1 subtypes was higher than that of other subtypes(P < 0.05). vacA m1 subtypes were closely related to the expression of MEX3A(P < 0.05), where MEX3A expression significantly correlated with the degree of tumor differentiation(P < 0.05). Conclusion In the Liaoning province of China,vacA m1 is the dominant subtype of H.pylori-positive gastric cancer. vacA m1 expression is related to MEX3A expression, and this may be involved in posttranscriptional regulation and related cell signaling pathways.
