摘要： 目的 明确己糖激酶-3(HK3)在乳头状肾细胞癌中的表达水平,探讨其与乳头状肾细胞癌患者的临床病理特征的相关性及对预后的影响。方法 通过TCGA数据库及Western blotting法检测HK3在乳头状肾细胞癌组织及癌旁组织中转录水平以及蛋白水平的表达差异。采用χ²检验分析HK3蛋白的表达情况及其与临床病理特征之间的关系;采用Kaplan-Meier法及log-rank检验绘制生存曲线并比较生存差异,分析HK3蛋白表达与无病生存期(DFS)和总体生存期(OS)的关系。结果 HK3在乳头状肾细胞癌组织中表达上调,差异有统计学意义(P<0.001)。HK3蛋白的表达水平与肿瘤的临床分期、Fuhrman核分级有关(P<0.05),与患者的年龄、性别及淋巴结转移无关(P>0.05);高表达的HK3与乳头状肾细胞癌患者的DFS及OS有关,差异有统计学意义(P<0.05)。结论 乳头状肾细胞癌组织中HK3的表达明显高于癌旁组织。HK3的高表达常提示患者的不良预后,筛查HK3的表达量对患者的临床治疗及预后具有一定的参考意义。

Abstract: Objective To investigate the expression of hexokinase 3 (HK3) in papillary renal cell carcinoma (pRCC) and explore its relationship with clinicopathological features and prognosis in patients with pRCC. Methods Differences in the transcriptional and translation levels of HK3 in pRCC and normal tissues were detected using The Cancer Genome Atlas database and Western blotting. The χ² test was used to analyze the relationship between HK3 protein expression and clinicopathological features,and the Kaplan-Meier and logrank tests were used to illustrate survival curves and compare survival differences. From these tests,the relationship between HK3 protein expression and disease-free survival (DFS) and overall survival (OS) was analyzed. Results HK3 expression was significantly upregulated in pRCC than in normal tissues (P< 0.001). HK3 protein expression level was correlated with tumor clinical stage and Fuhrman nuclear grade (P< 0.05) but not with age,sex,and lymph node metastasis (P> 0.05). Furthermore,high HK3 expression was significantly associated with DFS and OS in patients with pRCC (P< 0.05). Conclusion HK3 expression in pRCC was significantly higher than that in normal tissues. High HK3 expression often indicates poor prognosis. Therefore,screening HK3 levels has certain significance for assessing the clinical treatment and prognosis of patients.
