环状RNA hsa_circ_0009043在口腔鳞状细胞癌中的表达及临床意义

摘要: 目的 探讨hsa_circ_0009043在口腔鳞状细胞癌(OSCC)中的表达及其临床意义。方法 采用实时定量PCR测定hsa_circ_0009043在OSCC细胞系及组织中的表达水平,并分析其与临床病理特征的相关性。用受试者操作特征(ROC)曲线评估hsa_circ_0009043在OSCC诊断中的价值。基于生物信息学分析预测hsa_circ_0009043的潜在功能。结果 hsa_circ_0009043在OSCC细胞系及OSCC组织中的表达水平低于癌旁组织及正常口腔角质细胞(均P<0.05),其表达水平与肿瘤大小及淋巴结转移有显著相关性(P=0.01,P=0.037)。ROC曲线下面积为0.6769。基因本体(GO)和京都基因与基因组百科全书(KEGG)富集分析及hsa_circ_0009043/miRNA/mRNA调控网络显示,hsa_circ_0009043与OSCC进展存在一定相关性。结论 hsa_circ_0009043可能成为一种OSCC的生物标志物,并参与OSCC的发生、发展。

Abstract: Objective To investigate the expression of circular RNA hsa_circ_0009043 in oral squamous cell carcinoma (OSCC) and its clinicopathological significance. Methods qRT-PCR was used to detect the expression level of hsa_circ_0009043 in OSCC cell lines, OSCC tissue,adjacent tissue,and normal cells. The correlations between hsa_circ_0009043 expression and pathological characteristics were analyzed. Receiver operating characteristics (ROC) curve and database data were used to analyze the potential function of hsa_circ_0009043. Results The expression level of hsa_circ_0009043 in OSCC cell lines and tissues was lower than that in adjacent tissues and normal cells (HOK) (all P< 0.05). The expression level of hsa_circ_0009043 was significantly correlated with TNM stage and lymph node metastasis (P=0.01,P=0.037). The area under the ROC curve was 0.676 9. Gene Ontology and Kyoto Encyclopedia of Genes and Genomes enrichment analysis and hsa_circ_0009043/miRNA/mRNA regulatory network revealed the correlation of hsa_circ_0009043 with OSCC progression. Conclusion hsa_circ_0009043 may be a biomarker of OSCC and participate in the occurrence and development of OSCC.
