大黄酸通过Wnt/β-catenin信号通路抑制喉癌细胞的增殖、迁移和侵袭

Abstract: Objective To investigate the effects of rhein on the proliferation,invasion,and migration of Hep-2 cells in laryngeal carcinoma and its mechanism. Methods Laryngeal carcinoma Hep-2 cells were treated with low,medium,and high concentrations of rhein (6,12,and 24 μmol/L),and the proliferation,migration,and invasion capabilities of Hep-2 cells were detected by MTT,scratch,and Transwell assays. The expression levels of β-catenin,Dvl2,GSK-3β,and p-GSK-3β in Hep-2 cells were detected by quantitative real-time PCR and Western blotting. Results As the rhein concentration increased,the rates of proliferation,migration,and invasion of Hep-2 cells were significantly reduced (P< 0.05). Rhein downregulated the expression levels of Dvl2,p-GSK-3β,and β-catenin in Hep-2 cells (P< 0.05) and increased GSK-3β expression (P< 0.05). Activation of the Wnt/β-catenin signaling pathway by SKL2001 reversed the inhibitory effect of rhein on the migration and invasion of Hep-2 cells (P< 0.05). Inhibition of the Wnt/β-catenin signaling pathway by XAV939 inhibited the expression levels of Dvl2,p-GSK-3β,and β-catenin (P< 0.05) and further enhanced the inhibitory effect of rhein on the migration of Hep-2 cells (P< 0.05). Conclusion Rhein could effectively inhibit the proliferation,invasion,and migration of Hep-2 cells through the inhibition of the Wnt/β-catenin signaling pathway.
