Abstract: Objective To investigate the mechanism by which bone morphogenetic protein(BMP) receptor activin A receptor type 1 (ACVR1) regulates odontoblast polarity and promotes dentin formation in mice. Methods The Acvr1 gene was knocked out in mouse dental mesenchymal cells using the Cre-loxP system, by activating Cre recombinase under control of the Osterix promoter. Mandibular incisors were collected at postnatal day 21, and odontoblasts and dentin morphologies were observed by histology. Immunofluorescence staining was used to detect localization of polarity related markers and Golgi apparatus in odontoblasts. Dentin collagen formation was assessed by sirius red staining. Results Upon Acvr1 knockout, the boundary between mineralized dentin and predentin, and the boundary between predentin and the odontoblast layer displayed a zigzag pattern. Osteodentin was formed at the cusp, and the high columnar morphology of odontoblasts was lost, with abnormal localizations of markers for intercellular tight junctions and apical polarity associated protein. The position of the Golgi apparatus relative to the nucleus was also changed. The arrangement of collagen in dentin was irregular, and collagen maturity was decreased. Conclusion ACVR1 in dental mesenchyme promotes dentin formation by promoting odontoblast polarization in mice.
