Abstract: Objective To investigate the expression and localization of lipocalin-2 (LCN2) in breast cancer and its role in invasion and metastasis. Methods The expression and localization of LCN2 in breast cancer cell lines was detected by western blotting and immunohistochemistry and laser confocal microscopy, respectively. T47D breast cancer cells overexpressing LCN2 were cultured and detected by markers E-cadherin, claudin-1, vimentin, and slug. The effect of LCN2 on the migration of T47D breast cancer cells was detected by transwell. Results The expression of LCN2 was low in the MCF-7, T47D, and ZR-75-30 breast cancer cell lines;medium in the MDA-MB-231 breast cancer cell line;and high in the ZR-75-1 breast cancer cell line. Immunohistochemistry and laser confocal scanning microscopy demonstrated that LCN2 was localized in the cytoplasm. Western blotting analysis showed that the expression of E-cadherin and claudin-1 was significantly decreased in the T47D cells overexpressing LCN2, while the expression of slug and vimentin was significantly increased. The migration of the T47D cell line overexpressing LCN2 was promoted. Conclusion LCN2 is expressed at different levels in different breast cancer cell lines. It is mainly located in the cytoplasm and promotes migration of breast cancer cells. LCN2 expression contributes to epithelial-mesenchymal transition development in breast cancer cells.
