Abstract: Objective To investigate the expression of peroxiredoxin 2 (Prx2) in renal tissues of unilateral ureteral obstruction (UUO)rats. Methods Twenty-four male SD rats were randomly divided into sham (n=8) and UUO (n=16, rat renal fibrosis model was established) groups. Rats in sham group were sacrificed on day 3 post-surgery, and those in UUO group were sacrificed on days 3, 7, 14 and 21 post-surgery, respectively, and then, kidney tissues were harvested. HE staining was performed to evaluate the rat renal fibrosis model. The expression of Prx2 was detected through immunohistochemical analysis. The mRNA and protein expressions of Prx2, α-SMA, and Vimentin were detected through real-time PCR and western blotting. Results Compared with sham group, UUO group exhibited renal interstitial inflammatory cell infiltration, dilated renal tubules, increased collagen deposition, and eventually, renal fibrosis model was established successfully. The mRNA and protein expression levels of Prx2, α-SMA, and vimentin in UUO group were found to be increased with time (all P < 0.01). Conclusion Prx2 expression increased in renal tissues of UUO rats in a time-dependent manner, and the increase in its expression may be closely related to the degree of renal fibrosis.
