肺腺癌预后不良的重要基因CDC20、CCNB2和CDC45

Abstract: Objective To identify genes and potential mechanisms related to the prognosis of lung adenocarcinoma (LUAD). Methods Gene chips GSE33532,GSE32863,and GSE43458 were selected from the Gene Expression Omnibus database. GEO2R and Venn diagram were used to screen differentially expressed genes. The Database for Annotation,Visualization,and Integrated Discovery (DAVID) was used to perform Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) analyses. A protein-protein interaction (PPI) network was constructed by STRING and Cytoscape,and MCODE was used for module analysis. Kaplan-Meier Plotter was used for prognostic analysis. Gene Expression Profiling Interactive Analysis (GEPIA) was used to validate the differential expression of genes between LUAD and normal lung tissues. Results A total of 331 common differentially expressed genes were obtained,including 68 down-regulated genes and 263 up-regulated genes. Thirteen genes were significantly correlated with the prognosis of LUAD and were highly expressed in LUAD. Among them,3 genes (CDC45,CCNB2,and CDC20) are significantly enriched in the cell cycle pathway,and significantly affect the prognosis of different clinical and pathological factors of lung cancer patients. Conclusion CDC45,CCNB2,and CDC20 are related to poor prognosis of LUAD and are potential therapeutic target genes for lung adenocarcinoma.
