Abstract: Objective To explore the expression and clinical significance of interleukin-9 (IL-9) and interleukin-9 receptor (IL-9R) in non-small cell lung cancer tissues. Methods Between January 2017 and December 2018,paraffin-embedded specimens were prepared from cancerous and adjacent normal tissues from 110 cases of pathologically diagnosed non-small cell lung cancer,and fresh specimens of cancerous and surrounding normal tissue were collected from 19 non-small cell lung cancer patients,by surgical resection in the Department of Thoracic Surgery,Shengjing Hospital of China Medical University. Clinical data including gender,age,tumor differentiation degree,tumor size,lymph node metastasis,and TNM staging was collected for all patients. Immunohistochemistry was used to detect IL-9 and IL-9R expression in lung cancer tissues and adjacent tissues. Western blotting was used to detect IL-9 protein expression in lung cancer tissues and adjacent tissues. Correlations between IL-9 and IL-9R expression and tumor clinicopathology were assessed by χ2 analysis. Student's t test was used to compare IL-9 expression in non-small cell lung cancer tissues and adjacent tissues. Results By immunohistochemistry,expression of IL-9 and IL-9R were seen to be significantly higher in lung cancer tissues than in normal tissues adjacent to cancers (P<0.05). Western blotting results showed that IL-9 protein expression was significantly higher in lung cancer tissues than in normal tissues adjacent to cancers (P<0.05). IL-9 expression levels correlated with tumor size,lymph node metastasis,and TNM stage (all P<0.05),and IL-9R expression correlated with the patient's TNM stage and lymph node metastasis (all P<0.05). Conclusion IL-9 and IL-9R are highly expressed in non-small cell lung cancer tissues,and their expression levels correlate with tumor size,lymph node metastasis,and TNM stage. IL-9/IL-9R may represent a novel target for diagnosis and treatment of non-small cell lung cancer.
