circZEB1通过竞争性结合hsa-miR-656调控胃癌细胞的增殖和凋亡

Abstract: Objective To study the effect of circZEB1 on the proliferation and apoptosis of gastric cancer cells and decipher its underlying regulatory mechanism. Methods Nucleoplasmic separation experiments were used to detect the localization of circZEB1 in gastric cancer cells. Real-time PCR was used to detect the expression of circZEB1 in five gastric cancer cell lines and a physiologically epithelial cell line. CCK-8 assays and flow cytometry were used to detect the effect of circZEB1 on the proliferation,and the apoptosis and cycle distribution of gastric cancer cells,respectively. Fluorescein plum reporter assays were employed to verify the direct physical interaction between circZEB1 and hsa-miR-656. Results circZEB1 is mainly expressed in the cytoplasm of gastric cancer cells in addition to being highly expressed in gastric cancer cell lines. Knockout of circZEB1 in gastric cancer cells significantly inhibits the proliferation,promotes the apoptosis,and induces cell cycle arrest at G0/G1 phase. The fluorescein plum reporter assays confirmed that circZEB1 directly targets hsa-miR-656. Conclusion circZEB1 regulates the proliferation and apoptosis of gastric cancer cells by competitively binding to hsa-miR-656.
