Abstract: Objective To explore the effect of miR-149-5p on the migration of osteosarcoma cells. Methods The expression of miR-149-5p in osteosarcoma cells was detected using PCR. An miR-149-5p-overexpressing cell line was constructed using transfection technology. The expression of miR-149-5p was detected using PCR,and the expression of high mobility group protein A2 (HMGA2) was detected through western blotting. The migration of cells under different conditions was detected using the Transwell assay. The binding sites of miR-149-5p and the HMGA2 mRNA were predicted through the bioinformatic website StarBase. Results Compared to normal cells,osteosarcoma cells exhibited a lower expression of miR-149-5p (P<0.05) and higher expression of HMGA2 (P<0.05). miR-149-5p inhibited cell migration,which could be restored by HMGA2 overexpression. The bioinformatic data revealed that the HMGA2 mRNA can bind to miR-149-5p. Conclusion miR-149-5p regulates the migration of osteosarcoma cells through HMGA2.
