百令胶囊对支气管哮喘小鼠的治疗效果

摘要: 目的 探讨百令胶囊对支气管哮喘小鼠气道炎症、气道高反应和气道重塑的治疗效果。方法 将48只雌性Balb/c小鼠按随机数字表法分为正常对照组、支气管哮喘组、布地奈德组、低剂量百令胶囊组、中剂量百令胶囊组、高剂量百令胶囊组,每组8只。测定各组小鼠的气道阻力,计数小鼠肺泡灌洗液(BALF)中细胞总数。HE、AB-PAS和Masson染色观察气道炎症、气道重塑。采用免疫组化染色评估各组小鼠肺组织内α-平滑肌肌动蛋白(α-SMA)水平。结果 与支气管哮喘组相比,布地奈德组、低剂量百令胶囊组、中剂量百令胶囊组、高剂量百令胶囊组小鼠气道高反应、BALF中炎症细胞总数均降低(P<0.05);肺组织气道周围炎症减轻,杯状细胞化生降低,胶原沉积减少;α-SMA表达水平降低。低剂量百令胶囊组、中剂量百令胶囊组、高剂量百令胶囊组与布地奈德组比较,气道反应性无统计学差异(P>0.05)。除气道反应性外,布地奈德组上述指标均优于低剂量百令胶囊组、中剂量百令胶囊组、高剂量百令胶囊组。结论 百令胶囊可以抑制哮喘小鼠气道炎症、气道高反应和气道重塑。

Abstract: Objective To investigate the effects of Bailing capsule on the airway inflammation,remodeling,and resistance of mice with asthma. Methods 48 Balb/c mice were randomly divided into a control group,asthma group,budesonide group,and low-,middle-,and high-dose Bailing groups (n=8 per group). The airway resistance was measured,and total cells in the bronchoalveolar lavage fluid (BALF) were counted. HE,AB-PS,and Masson staining were performed to assess airway inflammation and remodeling. Immunohistochemical staining was performed to determine the expression of α-smooth muscle actin (α-SMA). Results Compared with the asthma group,the airway resistance and the number of total inflammatory cells in BALF decreased in the budesonide group and in the low-,middle-,and high-dose Bailing groups (P<0.05). Meanwhile,the inflammatory cell infiltration,goblet cell metaplasia,collagen deposition,and α-SMA expression in lung tissue also decreased. No significant difference in airway resistance was found between the budesonide and Bailing groups (P>0.05). Except for airway resistance,the aforementioned effects in the budesonide group were better than those in all of the Bailing groups. Conclusion Bailing capsule can inhibit airway inflammation,resistance,and remodeling in asthmatic mice.
