Abstract: The enzyme,nicotinamide N-methyltransferase (NNMT),methylates nicotinamide (NAM) to form N1-methylated NAM. Recent studies have shown that NNMT not only clears NAM but also participates in the regulation of various metabolic pathways,such as those in adipose and liver tissues,by consuming methyl donors and producing active metabolites. Herein,the roles of NNMT under physiological and pathological conditions and new therapeutic interventions are reviewed.
