虾青素对过氧化氢诱导的骨髓间充质干细胞氧化应激的保护作用

Abstract: Objective To investigate the effect of astaxanthin on hydrogen peroxide (H2O2)-induced oxidative stress response and the possible mechanism of that response in bone marrow mesenchymal stem cells (BMSCs). Methods Human BMSCs were cultured in vitro. H2O2 was used to induce oxidative stress in BMSCs. BMSCs were pretreated with varying concentrations of astaxanthin. Cell survival was analyzed via a CCK-8 assay,and cell apoptosis was detected using flow cytometry. A biochemical method was used to measure glutathione and malonyldialdehyde levels as well as superoxide dismutase activity. Western blotting was employed to determine nuclear factor erythroid-2-related factor 2 (Nrf2),heme oxygenase 1,and quinone oxidoreductase 1 expression levels. Results Astaxanthin significantly improved the survival rate of BMSCs that had been subjected to H2O2-induced oxidative stress;it also increased superoxide dismutase activity,increased glutathione level,and reduced malondialdehyde level. Furthermore,astaxanthin lowered the cell apoptosis rate and induced Nrf2 protein expression. Conclusion Astaxanthin may attenuate H2O2-induced oxidative stress in BMSCs by activating the Nrf2/antioxidant response element pathway.
