Abstract: Objective To screen the prognostic factors of stageⅠ gastric cancer (SIGC) by weighted gene co-expression network analysis (WGCNA) and to explore the molecular mechanisms in pathogenesis. Methods Clinical data from patients with SIGC in The Cancer Genome Atlas (TCGA) were analyzed by WGCNA. Survival analysis was performed to identify genes affecting SIGC prognosis. Additionally,the effect of this gene on prognosis was confirmed by cytological experiments. We further explored the mechanism by which this gene affected the prognosis of gastric cancer (GC) through gene set enrichment analysis (GSEA). Results We analyzed 330 samples from TCGA,which included 51 SIGC samples. Overall survival in patients with high expression of brain acid soluble protein 1 (BASP1) was lower than those with low expression of BASP1 (P=0.016 25). Cytological analysis showed that the migration ability of GC cell lines after knockdown of BASP1 decreased by 62.2%. GSEA indicated that the IL6-JAK-STAT3 pathway was highly enriched (NOM p-val < 0.001;FDR q-val < 0.001). Conclusion BASP1 is a marker for poor prognosis in SIGC,and it may affect GC cell migration through regulation of the IL6-JAK-STAT3 pathway.
