DDB2基因不同转录本在胃癌和正常胃组织中的表达差异及其对胃癌细胞增殖的影响

摘要: 目的 探讨DDB2基因不同转录本在胃癌和远端正常胃组织中的表达差异及其对胃癌细胞增殖的影响。方法 提取22对胃癌和远端正常胃组织的总RNA,利用实时定量PCR(RT-qPCR)检测DDB2基因WT和D1转录本的表达。构建DDB2基因WT和D1过表达质粒以及空载体,分别将其转染入胃癌BGC823细胞中。采用Western blotting检测各组细胞中转染质粒的表达情况、细胞凋亡行为和上皮-间充质转化情况,采用CCK-8实验检测各组细胞的增殖情况。结果 胃癌组织中DDB2基因D1转录本的表达水平显著高于远端正常胃组织(P < 0.01),而WT转录本的表达无明显差异。体外实验中,WT转录本促进BGC823细胞增殖(P < 0.05),而D1转录本抑制BGC823细胞增殖(P < 0.01)。2种转录本对胃癌细胞的凋亡行为和上皮-间充质转化现象无明显影响。结论 DDB2基因D1转录本在胃癌组织中表达增强,不同转录本对胃癌细胞的增殖行为产生相反作用。

Abstract: Objective To study the expression of different transcripts of the DDB2 gene in gastric cancer and distal normal gastric tissues,and their effects on the proliferation of gastric cancer cells. Methods Total RNA from 22 pairs of gastric cancer tissues and distal normal gastric tissues was extracted. The expression of DDB2 gene transcripts WT and D1 in the gastric cancer and distal normal gastric tissues were detected by quantitative real-time PCR. WT and D1 overexpression plasmids and empty vector were constructed and transfected into BGC823 cells. Western blotting was used to detect the expression of the plasmid,apoptosis behavior,and epithelial-mesenchymal transition. The proliferation of BGC823 cells was detected using the CCK-8 assay. Results The content of D1 transcript in gastric cancer tissues was significantly higher than that in distal normal gastric tissues (P < 0.01),while no significant difference in the content of WT transcript was found. In vitro,WT transcript promoted the proliferation of BGC823 cells (P < 0.05),while D1 transcript inhibited the proliferation of BGC823 cells (P < 0.01). There was no significant difference in the effects of these two transcripts on cell apoptosis and epithelial-mesenchymal transition. Conclusion Enhanced expression of DDB2 D1 transcript was found in gastric cancer tissues. Different transcripts displayed opposite function in the regulation of gastric cancer cell proliferation.
